VIRAL SELF-ASSEMBLY


VIRAL, SELF-ASSEMBLY
We have always been intrigued by the varied viral geometrical shapes, their location and meaning in the general context of living organic forms. Perhaps, the viruses are potential life, frustrous organic life or remains of organic life. Having been the ancestral viruses -for sure- extremely fragile -because of the second law of thermodynamics- it is logical to think that their old string of RNA or DNA, needed protective covers, as close as possible to microspheres in order to protect its genomic material and facilitate its insertion in host cells. As these small strings of ancestral RNA could not code spheres, it was opted for codes to form serial triangles, pentagons or hexagons, more affordable to be formed by organic elements, which after tessellation formed capsids like spheres. Fact that worked well until the formation of icosahedron viral capsids (20 faces), effective protectors of genomes with short RNAs, but not for viruses with larger RNA strings, as in the case of cancer-inducing viruses (polyomas, papillomas), with need for perfect tessellations and more complex and sophisticated geometric pairings. And so, as the Riemann mathematics helped Einstein understand that a geodesic is the shortest line in space, the construction of viral capsids using geometric shapes turned out to be also the simplest way to build effective genomic protective covers, capable of functioning and evolve according to circumstances. It must be understood -as several scientists have pointed out- that life is an emergent form of the evolution of matter, given certain environmental conditions. In a more mental tan practical work, which involves: mathematics-biology-mathematics, English scientists: Reidun Twarock (University of York/England) and Peter Stockley (University of Leeds), applied mathematical principles: geometry, Hamilton way, Penrose tilings, to understand RNA encodings, viral capsid formation, evolution and functioning of viruses, works that could open to the   english researchers the doors to a future Nobel Prize. Building on previous work by Watson, Crick, Franklin, Penrose Caspar-Klug and others, Twarock and Stockley have outlined the: a-Possibility of creating new and more effective antivirals and, b-Immunize millions of people against hundreds of viral diseases. After understanding Twarock, that mathematics would allow them to understand how viruses with larger RNA or DNA (polyoma and papilloma) produce effective capsids, she resorted to the use of group theory and discrete mathematics,  identifying first the positions of the protein subunits of the capsid of the human papilloma virus ordered in pentagonal structures like the Penrose tiles that do not repeat periodically, produce large RNA strings and leave no spaces when 2 subunits are joined. According to Twarock, there are remnants of that ancestral order in simple viral genomes. The scientist says that the viral genomic material plays a very active role in the formation of capsids, mentioning with emphasis the signals of viral packaging (contact areas of the walls of the capsid with the genome), which help the viral assembly, the same ones that are located according to the guidelines of the Hamilton way and whose current identification would certify the conservation of a primitive genomic organization. The viral packaging signals allow a protein to adhere to them, to which other proteins will later adhere forming orderly pathways that never repeat themselves (Hamilton way). The attack with antivirals to these signals would prevent the formation of capsids. For his part, Peter Stockley, tries to create stable non-infectious immunogens: vaccines able to induce the formation of antibodies without producing side effects. Twarock tries to use the viral packaging signals and the subsequent self-assembly to make synthetic RNA and viruses, unable to replicate, but generating an immune response.

AUTOENSAMBLAJE, VIRAL


Siempre nos han intrigado las variadas formas geometricas virales y su ubicación y significado en el contexto general de las formas organicas vivientes. Quizas, los virus sean vida potencial, ensayos frustros o restos de formas organicas de vida. Habiendo sido los virus ancestrales -con seguridad- extremadamente frágiles -en razón de la segunda ley de la termodinámica- es lógico pensar que sus antiguas cuerdas cortas de RNA o DNA, necesitasen cubiertas protectoras semejantes, lo mas cercanamente posibles a microesferas a fin de proteger su material genómico y facilitar su inserción en celulas huésped.  Como estas pequeñas cuerdas de RNA ancestral no podían codificar esferas, optaron por  codigos para formar triangulos, pentágonos o hexágonos en serie -mas asequibles a ser formadas por elementos organicos-  los que tras teselarse formaron cápsides semejantes a esferas. Hecho que funciono bien hasta la formación de cápsides virales icosahedricas (20 caras), protectoras eficaces de genomas con RNA cortos, pero no para virus con cuerdas de RNA, mas grandes, como el caso de  virus inductores de cáncer (poliomas, papilomas), con necesidad de teselados perfectos y emparejamientos geométricos mas   complejos y sofisticados. Y asi, como las matematicas de Riemann ayudaron a Einstein a entender que una geodésica es la línea mas corta en el espacio, la construcción de cápsides virales empleando formas geométricas resulto ser también la forma mas simple de construir cubiertas protectoras genómicas eficaces, capaces de funcionar y evolucionar de acuerdo a las circunstancias.  Debe entenderse -como lo han señalado varios científicos- que la la vida es una forma emergente de la evolucion de la materia, dadas ciertas condiciones medioambientales. En un trabajo mas mental que practico, que involucra:  matemática-biología-matematica, los científicos ingleses:  Reidun Twarock (University of York/England) y Peter Stockley (University of Leeds), aplican  principios matemáticos: geometría, algoritmos de Hamilton, geometría de Penrose, para comprender las codificaciones de RNA, formación de cápsides virales, evolución y funcionamiento de los virus, trabajos que podrían abrirles las puertas a un futuro Premio Nobel. Apoyandose en trabajos previos de Watson, Crick, Franklin, Penrose Caspar-Klug y otros, Twarock y Stockley han esbozado la: a-Posibilidad de crear nuevos y mas eficaces antivirales y, b-Inmunizar a millones de personas contra cientos de enfermedades virales. Tras entender Twarock, que las matematicas les permitirían  entender como virus con RNA o DNA mas grandes (polioma y papiloma), producen  cápsides eficaces, recurrio al empleo de la teoría de grupos y matematicas discretas, identificando primero las posiciones de las subunidades proteicas de la cápside del virus del papiloma humano ordenado en estructuras pentagonales al modo de las losetas de Penrose que no se repiten periódicamente, producen grandes cuerdas de RNA  y  no dejan espacios cuando se unen 2 subunidades. Según Twarock, existen remanentes de de ese orden ancestral en genomas virales simples.  La científica, afirma que el material genómico viral juega un rol muy activo en la formación de cápsides, mencionando con énfasis a las señales de empaquetamiento viral (áreas de contacto de las paredes de la cápside con el genoma), que ayudan al ensamblaje viral, las mismas que se ubican según  los lineamientos  de la via de  Hamilton y cuya identificación actual certificaria la conservación de una  organización genómica primitiva. Las señales de empaquetamiento viral permiten que una proteína se adhiera a ellas, a la que posteriormente se adherirán otras proteínas formando vías ordenadas que nunca se repiten (Via de Hamilton). El ataque con antivirales a estas señales impediría la formación de capsides.  Por su lado, Peter Stockley, intenta crear   inmunógenos estables no infecciosos: vacunas capaces de inducir la   formación de anticuerpos sin producir efectos colaterales. Twarock intenta emplear las señales de empaquetamiento viral y el subsecuente autoensamblaje para fabricar virus y RNA sintéticos, incapaces de replicarse, pero generando una respuesta inmune.

EXTENDED HUMAN BEING LIFE

RECYCLED ANTICANCER GENES
We thought that prolongation of  human  useful life would be linked to human body recycling with embryonic stem cells or, to an enhancement of the immune system -that is- eliminating harmful noxious, would extend human life. Today, we have a novel, simple, extraordinary and improved proposal, proven again and again by the evolution of the species. The fossil remains of elephants (30 million years ago), when they struggled to acquire giant body conformations -showed a paradox- large bodies with prolonged lives are exposed to developing neoplasms. Therefore, Richard Peto (cancer epidemiologist/University of Oxford) was surprised that the large size of the elephants were not associated with high cancer frequencies. The low frequency of neoplasms (3%), in elephants, was understandable only if, shortly before acquiring large sizes, these animals had converted a portion of their genetic material junk into a defense against cancer. One hypothesis suggested that an old non-functional gene had been recycled from the abundant genomic junk, increasing the sensitivity of   incipient tumor cells to an early death (apoptosis). In 2015, J. Schiffman (Utah School of Medicine and Carlo Maley (Arizona State University), showed that  elephant's genome had 20 extra duplicates of the suppressor gene (p53), suggesting that the function of these copies had a cancer  suppressive effect. Vincent Lynch(University of Chicago), discovered that one of these oncogenes: LIF6 (a non-functional duplicate of an ancestral gene), had suddenly adopted an anti-cancer function. According to Lynch, elephants and their small relatives (armadillos, hyraxes, and aardvarks), also duplicate copies of their LIF gene (leukemia inhibitor factor), noting that normally this protein is related to fertility, reproduction and growth stimulation of embryonic stem cells. None of these duplicates could carry out normal gene functions.  In 2017, Lynch showed that 11 duplicates of LIF gene were incomplete (lacked the initial block of  coding protein of information and the promoter sequence to regulate the activity of the gene). Lynch found RNA transcripts in a LIF duplicate, which had a promoter sequence of a few thousand bases of LIF6 and a DNA sequence similar to a binding area for the p53 protein, suggesting that p53 regulated the expression of LIF6. The LIF6 gene blocked the activity of the gene and prevented apoptosis (a control to eliminate defective tissue). The LIF 6, eradicator of malignant cells, created breaks in the surrounding membranes of the mitochondria. Lynch also found in genomes of closely related species (manatees, hyraxes, extinct mammoths, mastodons), evidence that LIF 6 gene had been duplicated 17 times and lost 14, during the evolution of the lineage of the elephants. Hyraxes and manatees had duplicates of LIF6, but duplicates of p53 only appeared in live and extinct elephants. Most duplicates of LIF gene are pseudogenes: copies of unused genes that survived in genome at random. LIF 6 is a re-functionalized pseudogene that re-evolved into a functional gene, starting from an ancestor and only modern and extinct elephants possess it, since they diverged from their related species. Not all animals have a re-functionalized LIF 6, because this protection has risks. LIF6 suppresses cancer, but extra copies of LIF6 could kill normal cells if the copies were accidentally activated. The LIF is critical for the implantation of the embryo in the uterus. The LIF and p53 together regulate the efficiency of reproduction. When the same set of genes has 2 functions (reproduction and suppression of cancer), they come into conflict (antagonistic pleiotropy). The elephants resolved this antagonism by duplicating their p53 and LIF genes and dividing their functions. According to Maley, some copies of p53 and LIF are dedicated to fertility, while other pairs of LIF and p53 suppress cancer. This method would not be a unique solution, because certain whales have a single copy of p53 and LIF. According to Maley, this knowledge will have applications in the prevention of cancer.
 GENES ANTICANCER RECICLADOS

Pensábamos que la prolongación de la vida útil humana estaría ligada al reciclaje corporal humano con células madre embrionarias o, a una potenciación del sistema inmune que, eliminando noxas perjudiciales, extendería la vida humana. Hoy, disponemos de una novedosa, sencilla, extraordinaria y mejorada propuesta, probada una y otra vez por la evolución de las especies. Los restos fósiles de elefantes de hace 30 millones de años, cuando pugnaban por adquirir conformaciones corporales gigantescas -muestran una paradoja-: los cuerpos grandes con vidas prolongadas están expuestos a desarrollar neoplasias. Por ello, A Richard Peto (epidemiólogo del cáncer/Universidad de Oxford), le sorprendió que el gran tamaño de los elefantes no se asociase a altas frecuencias de cáncer. La baja frecuencia de neoplasias (3%), en los elefantes, se hacía comprensible solo, si poco antes de adquirir grandes tamaños, estos animales hubiesen convertido una porción de su material genético basura, en una defensa contra el cáncer. Una hipótesis sugería que un gene antiguo no funcional, hubiese sido reciclado de la abundante basura genómica, incrementándose la sensibilidad de las células tumorales incipientes a una muerte temprana (apoptosis). El 2015, J. Schiffman (Utah School of Medicine y Carlo Maley (Arizona State University), demostraron que el genoma del elefante tenía 20 duplicados extras del gene supresor (p53), sugiriendo que la función de estas copias tuviese un efecto supresor del cáncer. Vincent Lynch: (Universidad de Chicago), descubrió que uno de estos oncogenes: LIF6 (duplicado no funcional de un gene ancestral), había adoptado de súbito una función anticancerígena.  Según Lynch, los elefantes y sus parientes pequeños (armadillos, damanes y cerdos hormigueros), también duplican copias de  su gene LIF (inhibidor de la leucemia), advirtiendo que normalmente esta proteína se relaciona con la fertilidad, reproducción y estimulación del crecimiento de células madre embrionarias. Ninguno de los duplicados podía llevar a cabo las funciones normales del gene.   El 2017, Lynch demostró que 11 duplicados del gene LIF, eran incompletos (les faltaba el bloque inicial de la proteína codificadora de la información y la secuencia promotora para regular la actividad del gene). Lynch encontró transcriptos de RNA en un duplicado LIF, que tenía una secuencia promotora de pocos miles de bases de LIF6 y, una secuencia de DNA semejante a un área de unión para la proteína p53, sugiriendo que el p53, regulaba la expresión   del LIF6.  El gen LIF6, bloqueaba la actividad del gene e impedía la apoptosis, un control para eliminar tejido defectuoso. El LIF 6 erradicador de células malignas, creaba roturas en las membranas circundantes de las mitocondrias. Lynch además encontró en genomas de especies estrechamente relacionadas (manatíes, damanes, mamuts extintos, mastodontes), evidencias de que el gene LIF 6 había sido duplicado 17 veces y perdido 14, durante la evolución del linaje de los elefantes. Damanes y manatíes tenían duplicados del LIF6, pero los duplicados del p53 solo aparecían en elefantes vivos y extintos.  La mayoría de los duplicados del gene LIF son pseudogenes: copias de genes sin uso que sobrevivieron en el genoma al azar. El LIF 6 es un pseudogene re-funcionalizado que re-evoluciono a un gene funcional, a partir de un ancestro y que solo lo poseen los elefantes modernos y extintos, desde que divergieron de sus especies relacionadas. No todos los animales tienen un LIF 6 re-funcionalizado, porque esta protección tiene riesgos. El LIF6 suprime el cáncer, pero las copias extras de LIF6 podrían matar células normales si las copias fuesen activadas accidentalmente. El LIF es crítico para la implantación del embrión en el útero. El LIF y el p53 regulan juntos la eficiencia de la reproducción. Cuando el mismo set de genes tiene 2 funciones (reproducción y supresión del cáncer), entran en conflicto (pleiotropia antagónica). Los elefantes resolvieron este antagonismo, duplicando sus genes p53 y LIF y dividiendo sus funciones. Según Maley, algunas copias de p53 y LIF se dedican a la fertilidad, mientras otros pares de LIF y p53 suprimen el   cáncer. Este método no sería una solución única, porque ciertas ballenas tienen una sola copia de p53 y LIF. Según Maley estos conocimientos tendrán aplicaciones en la prevención del cáncer.

¿CURE OF SCHIZOPHRENIA?

Well could Oliver Howes (Molecular Psychiatry/MRC London Institute of Medical Sciences), be one of the next winners of the Nobel Prize in Medicine. The long work done by him and his team during the last 10 years begins to give him, mankind and thousands of schizophrenics distributed throughout the World  revenues. After ascertaining Howes and his team, that the increase in the number and activity of  certain brain immune cells  (microglia), induce them to actively destroy unwanted neuronal connections, began they to study this finding   in depth. Great was the surprise of these researchers when they found that neuronal pruning fundamentally compromises the frontal area and auditory regions, explaining why schizophrenics have cognitive defects and hear voices. It is now known that the tissue scenarios of inflammation and psychotic activity induced by autoimmune mechanisms rapidly improve by reducing inflammation. Although traditional antipsychotic medications block the action of dopamine and improve schizophrenia a little, they do not protect the brain structure. Thanks to the collaboration between scientists of the MRC and King's College London, 60 patients will receive for 3 months the monoclonal antibody Natalizumab, used with relative success and since several years against multiple sclerosis. In the case of schizophrenia, the Natalizumab is expected to attack   microglía  cells, restricting its movements around the brain, thus preventing the pruning of vital neuronal connections. The first patient already enrolled will receive monthly infusions of Natalizumab, expected to heal or improve him. With these advances, an empowerment of biological psychiatry is expected, visualizing in the near future, the eradication of most mental illnesses. A new scientific frontis is expected: mind-body indivisibility, missing only to deepen more studies in this area. New doors open for the treatment of mental illness.


¿CURA DE LA ESQUIZOFRENIA?

Bien podría Oliver Howes (Psiquiatría Molecular/MRC London Institute of Medical Sciences), ser uno de los próximos ganadores del Premio Nobel de Medicina. El largo trabajo realizado por el y su equipo durante los últimos 10 años empieza a darle réditos a él, a la humanidad y a miles de esquizofrénicos distribuidos a lo largo y ancho de la Tierra. Tras cerciorarse Howes y su equipo, de que el incremento en el número y actividad de  células inmunes  cerebrales (microglía), las inducia  a  destruir activamente conexiones neuronales no deseadas, empezó a estudiarse a fondo este hallazgo. Grande fue la sorpresa de estos investigadores al constatar que la poda neuronal compromete fundamentalmente el área frontal y las regiones auditivas, explicando porque los esquizofrénicos tienen defectos cognitivos y escuchan voces. Hoy se sabe, que los escenarios tisulares de inflamación y actividad psicótica inducidos por mecanismos autoinmunes mejoran rápidamente al reducir la inflamación. Aunque los medicamentos antipsicóticos tradicionales al bloquear la acción de la dopamina mejoran algo la esquizofrenia, no protegen la estructura cerebral.  Merced a la colaboración entre científicos del MRC y el King’s College London, 60 pacientes recibirán durante 3 meses el anticuerpo monoclonal    Natalizumab, empleado con relativo éxito y desde hace varios años contra la esclerosis múltiple.  En el caso de la esquizofrenia, se espera que el Natalizumab ataque a las células microglía, restringiendo sus movimientos alrededor del cerebro, impidiendo así, la poda de conexiones neuronales vitales.  El primer paciente ya enrolado recibirá infusiones mensuales de Natalizumab, esperándose que se cure o mejore.  Con estos avances, se espera un empoderamiento de la psiquiatría biológica, visualizándose en el futuro próximo, la erradicación de la mayoría de las enfermedades mentales. Se espera un nuevo frontis científico:  indivisibilidad mente-cuerpo, faltando solo profundizar más estudios en este rubro. Se abren nuevas puertas para el   tratamiento de las enfermedades mentales.

ALPHA GO

The analysis of  several  hobbies, including the ancient Chinese strategy game Go, has allowed researchers of  Artificial Intelligence (IA) program from Google's Deep Mind to create Alpha Go, a strategic game, able to create their own rules, based on little related information and without any human intervention. Early versions of Alpha Go (Zero) employed trial and error and 2 neural networks (imitations of human neural networks): to predict the best movements and results, through quick and random games. The currently Alpha Go Zero, employs a single neural network, which is asked who won in a certain position (as you ask an expert). Resting in a single neural network has made this algorithm more efficient and powerful, although more computational power is required. One of the objectives of Deep Learning  in relation to Alpha Go is to analyze deeply the systems used by the human brain to solve problems. Now, Alpha Go, that  trains itself, employs strategies not created by humans in thousands of years, reaching superhuman levels in game strategy. Without the influence of emotions and other pressures, these machines are expected to outperform the human brain in logical-rational aspects. Therefore, its creators intend to use it to solve problems of robotics, construction of materials and assembly of protein folds. The first version: from Alpha Go, started from scratch rules, imitate the structure of the brain and played against  the best human players  to learn abstract concepts. Established the first rules, the ancestral versions of Go, increased each time, its information, improving after each game. During the first phase imitated what the best human players did (avid capture of the opponent's chips). Then, they used complex tactics used by human experts, accumulated over thousands of years, to eventually create new strategies and knowledge.

ALPHAGO


El análisis de varios pasatiempos, dentro de ellos el ancestral juego de estrategia chino Go, ha permitido  a  un programa de investigadores de  Inteligencia Artificial (IA), de   Deep Mind de Google, crear Alpha Go, un juego estratégico     capaz de crear sus propias reglas,  partiendo de poca información relacionada y sin mediar   intervención humana alguna. Las  primeras versiones de Alpha Go (Zero), empleaban el ensayo y el error y 2 redes neurales  (imitaciones de las redes neurales humanas): para predecir los mejores movimientos y otra para     evaluar los  resultados,mediante juegos  rápidos  y  al azar.    El Alpha Go Zero, actual emplea   una sola red neural, a la que se le  pregunta quien  ganara en determinada posición (como se le pregunta a un experto). Reposar en una sola red neural ha convertido a este algoritmo en más eficiente y poderoso, aunque se requiere mayor poder computacional. Uno de los objetivos de Deep    Learning en relación a Alpha Go, es analizar de profundis los sistemas que emplea el cerebro     humano para resolver problemas. Ahora, Alpha Go  se entrena a sí mismo,  emplea  estrategias  no creadas por humanos en miles de años, alcanzando niveles superhumanos en     la estrategia del juego. Se prevé que, sin la influencia de las emociones y otras presiones, estas máquinas superaran al cerebro humano en aspectos lógico-racionales. Por ello,  sus  creadores  pretenden emplearlo para resolver problemas de robótica,creación de materiales y     ensamblaje de pliegues de  proteínas. La    primera versión: de Alpha Go, empezó de cero      aprendiendo a jugar  contra sí mismo.   Tras vencer a los mejores jugadores del mundo y practicar técnicas de reforzamiento,el algoritmo directriz,  sobrepaso las versiones iniciales   empleando  menor tiempo de entrenamiento y mayor poder    computacional. Alpha Go emplea    imitaciones de redes neurales humanas inspiradas en la estructura del cerebro, para aprender   conceptos abstractos. Establecidas    las primeras reglas, las versiones ancestrales de Go, incrementaron cada vez, su información,   mejorando después de cada juego. Durante la   primera fase   imitaban lo que hacían los mejores jugadores humanos (ávida captura de fichas del contrario). Luego emplearon tácticas  complejas usadas   por expertos humanos,   acumuladas durante  miles           de años, para al final crear nuevas estrategias y    conocimientos.

PRIZE NOBEL IN PHYSICS,2017

The detection of gravitational waves  disruptors of time-space,  produced somewhere in the cosmos, 1.3 billion years ago as a result of a collision between two black holes, would apparently favor new ways of determining spatial mapping, size and age of the universe that  we inhabit, data from the Big Bang, dark matter, supernovae. etc. We say, apparently, because  75% of the known universe is dark energy and 25% is         occupied by   dark matter (where elementary particles may be different from known ones). So, it is too early to assess their perspectives. Although these waves were predicted 100 years ago, by Albert Einstein in his general theory of relativity, their detection for the first time was made on September 14, 2015, using the Laser Interferometer Gravitational-Wave Observatory (LIGO) detector. Given the enormous distance traveled, the intensity of the waves was very weak when they reached Earth. To detect them it was necessary to use giant     laser interferometers capable of measuring waves thousands of times smaller   than the atomic nucleus of any element. The screening allowed the award of the Nobel Prize in Physics, to Rainer Weiss(MIT, Cambridge, MA, USA), Kip S. Thorne (California Institute of Technology,Pasadena, CA, USA) and Barry C. Barish of Technology, Pasadena, CA, USA).

PREMIO NOBEL DE FÍSICA, 2017


La detección de ondas gravitacionales -disruptoras del espacio tiempo- producidas en algún lugar del cosmos, hace 1,3 billones de años a producto de una colisión entre dos agujeros negros, aparentemente favorecerían nuevas formas de determinar la cartografía espacial, del tamaño (las ondas viajan a la velocidad de la luz y llenan el cosmos) y edad del universo que habitamos, datos del Big Bang, de la materia oscura, de las supernovas. etc. Decimos aparentemente porque con un 75% del universo conocido copado por energía oscura           y un           25%  por  materia oscura (donde podrían existir partículas elementales diferentes a las conocidas),  es muy temprano para valorar sus perspectivas.  Aunque estas ondas, fueron predichas hace 100 años, por Albert Einstein en su teoría general de la relatividad, su detección por primera vez fue realizada el 14 de Setiembre del 2015, mediante el detector LIGO (Laser Interferometer Gravitational-Wave Observatory). Dada la enorme distancia recorrida, la intensidad de las ondas fue muy débil cuando estas alcanzaron la Tierra. Para detectarlas fue necesario emplear interferómetros gigantes capaces de medir ondas miles de veces más pequeñas que el núcleo atómico de cualquier elemento. La detección permitió la concesión del Premio Nobel de Fisica,2017 a: Rainer Weiss (MIT, Cambridge, MA, USA),  Kip S. Thorne (California Institute of Technology, Pasadena, CA, USA) y   Barry C. Barish (California Institute of Technology, Pasadena, CA, USA).

PRIZE NOBEL IN CHEMISTRY, 2017




NOBEL CHEMISTRY AWARD, 2017

Until recently, advances in biochemistry were possible by the realization and interpretation of biochemical reactions, but without adequate visualization of the resulting biomolecular structures. Until the invention of the electronic microscope (ME), that began to change the panorama, visibilizing clearly, some biostructures in 2 dimensions (2D), with a certain size limit. The overcoming of the above limitations allowed the award of the Nobel Prize in Chemistry, 2017, to JacquesDubochet (University of Lausanne, Switzerland), Joachim Frank (ColumbiaUniversity, NY) and Richard Henderson (MRC Laboratory of Molecular Biology,Cambridge, UK), by enabling many biomolecules to be seen at the atomic level in 3D, simplifying and improving the images and intuiting the function of many of them. Achievement achieved with the invention of cold cryo-microscopy of high resolution. An achievement conditioned by 3 master ideas: 1) Use of low intensity electron beams to not destroy biological samples. 2) Avoiding evaporation of water by cooling the biological samples rapidly in liquid methane or propane at -190 ° C, preserving it in liquid nitrogen and maintaining it in a liquid (amorphous) liquid state and with molecular weights greater than ~ 50 kDa and 3) generation of softwares that allowed to transform the atomic images of 2D in 3D. What's next? Although we do not know, we intuit that what comes is more technology; add more pixels to the images, better resolution to the  same,  clear observation of smaller images.

Hasta hace poco, los avances en Bioquímica fueron posibles por la realización e interpretación de reacciones bioquímicas, aunque sin visibilizar adecuadamente, las estructuras biomoleculares resultantes. Hasta que la invención del microscopio electrónico (ME), empezó a cambiar el panorama, visibilizándose con claridad, algunas bioestructuras en 2 dimensiones (2D) y con cierto límite de tamaño. La superación con creces de las limitaciones anteriores, permitió la concesión del Premio Nobel de Química, 2017, a Jacques Dubochet (University of Lausanne, Switzerland), Joachim Frank (Columbia University, NY) y
Richard Henderson (MRC Laboratory of Molecular Biology, Cambridge, UK), al posibilitar que muchas biomoléculas sean vistas a nivel atómico, en 3D, simplificando y mejorando las imágenes e intuyéndose la  función de muchas de ellas. Logro conseguido con la invención de la   crio-microscopia al frio, de alta resolución.  Un logro condicionado   por 3
ideas maestras: 1) Empleo de haces de electrones de baja intensidad para no destruir las muestras biológicas.  2) Evitando  la evaporación del agua enfriando las muestras biológicas rápidamente  en metano liquido o propano a  -190ºC, preservándola en nitrógeno líquido y,  manteniéndola en    estado líquido vitrificado (amorfo) y con pesos moleculares mayores de  ~50 kDa y 3) generación de softwares que permitieron  transformar  las imágenes atómicas de 2D en 3D. ¿Qué viene ahora?.Aunque no lo sabemos intuimos que lo que viene es más tecnología; -agregar más pixeles a las imágenes -mejor resolución de las mismas  – observación nítida de imágenes de menor tamaño

NOBEL PRIZE FOR MEDICINE AND PHYSIOLOGY, 2017

How to interpret the fact that all living organisms (plants, animals and humans), must to anticipate and adapt their physiologies to the sequential, environmental changes caused by the succession of day and night, which in turn induce adaptive body changes (higher or  lower temperature, lower or higher levels of hormones, blood pressure, alertness levels, or altered  behaviors)?. This is a necessary question in the case of studies that, over many years, have proved the existence  -at least until now- of 3 pairs of genes with their respective encoded proteins that allow the corporal adaptations referred  and that have allowed to  3 American scientists: Jeffrey C. Hall (Brandeis University, Waltham), Michael Rosbash (BrandeisUniversity) and Michael W. Young (Rockefeller University, NY), get the Nobel Prize for Medicine and Physiology, 2017, for their  discoveries of the molecular mechanisms that control the circadian rhythm. 1-An interpretation is that we -humans- are adaptive, autonomous, programmed biomachines, that we have biological plasticity and that free will, should be rethought. 2-If the environment influences us to the point of manipulating our genes, then, the medicine based on colors, and others related would have their reason for being. 3-Considering that some diseases are produced by defective genetic adaptations to the environment, then the care and improvement of the environment should be part of medicine.4-On the other hand, the genetic mechanism that generates proteins (that are elaborated at night and that are degraded during the day), and which promotes the adaptation of our physiology to environmental changes is an example of autonomous biological engines, which although not yet created by the human mind, are everywhere present in the human body.

¿Como interpretar el que todos los organismos vivos (plantas, animales y humanos), tengan que anticipar y adaptar sus fisiologías a los cambios secuenciales, medioambientales, provocados por la sucesión del día y la noche, inductores a su vez de cambios corporales adaptativos (mayor o menor temperatura, menores o mayores niveles de hormonas, de presión arterial, grados de alerta o, que sus conductas se alteren)?. Pregunta necesaria ante  estudios que a lo largo de muchos años han probado   la existencia -por lo menos hasta ahora- de 3 pares de genes con sus respectivas proteínas codificadas que permiten las adaptaciones corporales referidas y que han permitido que 3 científicos americanos: Jeffrey C. Hall (Brandeis University,Waltham), Michael Rosbash  (Brandeis University) y  Michael W. Young (Rockefeller University,NY), obtengan el Premio Nobel de Medicina y Fisiología, 2017, por  descubrimientos de los mecanismos moleculares que controlan el ritmo circadiano. 1-Una interpretación es que seamos biomaquinas adaptativas, autónomas, programadas, que tenemos plasticidad biológica y que el libre albedrio debiese ser repensado. 2-Si, el medio ambiente influye en nosotros al punto de manipular nuestros genes, entonces, la medicina basada en colores y otras  relacionadas, tendrían su razón de ser. 3-Considerando que algunas enfermedades son producidas por adaptaciones genéticas defectuosas al medio ambiente, entonces el cuidado y mejora del ambiente debiese formar parte de la medicina.4-De otro lado, el mecanismo genético que genera proteínas (que se elaboran  de noche y se degradan durante el día) y,  que promueven la adaptación de nuestra fisiología a cambios medioambientales es  ejemplo de motores biológicos autónomos, que aunque aún no han sido  creados  por la mente humana,  están presentes por doquier en el cuerpo humano.

PROXIMA CENTAURI b







NEXT GOAL :PROXIMA  CENTAURI b


In 2016, Stephen Hawking hypothesized to reach distant habitable stars in a few years, using space-based nanoprobes  mobilized at 1/5 the speed of light, propelled by laser beams equivalent to 100 billion watts, observing stars, photographing them and sending  collected  data to the Earth. Without wasting time, Russian entrepreneur Yuri Milner financed this project (Breakthrough Starshot), with 100 million dollars for the first 5 years. The successful launch of several of these nanosatelites (Sprites/3.5cm x 3.5cm, weight: 4 g, cost: 10 dollars), equipped with navigation equipment, cameras, communication systems, solar cells, radio sensors, computers), exceeds previous versions of minisatelites type Cube Sats (10 cm x 10 cm, weight: more than 1 kg). Given their mini-size, the Sprites intend to arrive at Proxima Centauri b, an exoplanet that orbits within the habitable zone of the red dwarf Proxima Centauri, the  nearest star to our Sun, distant 4.2 light years from Earth.  Sprite Project, led by Zac Manchester (Harvard University), has placed 2 Sprites on the outside of the Latvian Venta macro-satellite and 4 on the Max Valier, built by the German company OHB System AG (June, 2017), soon to be freed  into space, where they will explore the possible existence of water and life beyond Earth.

El 2016, Stephen Hawking hipotetizo alcanzar estrellas habitables lejanas en pocos años, empleando  nanosondas espaciales  movilizadas a  1/5 de la velocidad de la luz,   propulsadas por  rayos laser equivalentes  a 100 billones de vatios,  observandolas, fotografiandolas  y enviando  los datos recopilados a la Tierra. Sin perder tiempo, el empresario  ruso  Yuri Milner financio este proyecto (Breakthrough Starshot),  con 100 millones de dolares   para  los primeros 5 años. El lanzamiento exitoso de  varios de estos nanosatelites  (Sprites/3.5cm x 3.5cm,  peso : 4 g,  costo: 10 dolares), apertrechados con equipos de navegacion, camaras fotograficas, sistemas de comunicacion, celdas solares, radios sensores, computadoras),  supera las versiones previas  de minisatelites tipo   Cube Sats (10 cm x 10 cm, peso :mas de 1 kg). Dado su minitamaño, los Sprites pretenden arribar a   Proxima Centauri b,  un exoplaneta que orbita dentro de la zona habitable de la estrella enana roja Proxima Centauri, la estrella, más cercana a nuestro Sol,  distante 4,2  años luz de la Tierra. El Proyecto Sprite liderado por Zac Manchester (Harvard University), ha colocado  2 Sprites en   la parte externa del   macro-satelite   Latvian Venta y 4 en el Max Valier,  construidos por la compañia alemana  OHB System AG  (Junio,  2017), a fin de ser   liberados prontamente al  espacio, donde exploraran la posible existencia de agua y vida mas alla de la Tierra.

TREATING GLIOBLASTOMAS








PROLONGED REMISSIONS IN CEREBRAL TUMORS


Recent  news have reported the  possibility not only to get  prolonged remissions, but of healing of certain types of cancer, by simply copying and enhancing the normal cellular defenses of our own humanity (Shawn Hingtgen: Tumor-homing cytotoxic humaninduced neural stem cells for cancer. Therapeutic Medicine, Vol. 9 Issue 375). One more case of the correct use of natural medicine. 1) The article reports observations made in mice and humans with brain tumors (glioblastomas), in which autologous,  reprogrammed neural stem cells (NSCs), from adult  skin cells, migrate to tumor cells in response to signals (tumor homing properties), released by tumor cells, thereafter infiltrating primary and invasive foci of glioblastomas, reducing the size of tumor cells to 2% or 5% of their original size. A novel therapeutic, since surgery, chemotherapy or radiation are unable to eradicate recurrences of glioblastomas. 2) It is necessary to consider that it is difficult to obtain  autologous  NCSs,  and that NSCs from donors have the risk of rejection and other collateral events. Bagó J.R., bypassed  this risk using mouse skin fibroblasts to which he transdiferentiated  to NSCs, a process that in mice lasts 4 days. Once obtained, human NSCs are injected directly into brain   glioblastomas reducing 20 to 50 times their size, in 24-28 days. In humans, NSCs, in addition to infiltrating tumor cells, release into glioblastomas: cytotoxic therapy, gene products, proapoptotic substances, tissue necrosis alpha factor, etc., reducing 250 times the size of glioblastomas in 3 weeks (Methods 2016.Neural stem cell therapy for cancer).



REMISIONES PROLONGADAS EN TUMORES CEREBRALES

La siguiente    noticia encierra la posibilidad no solo de  lograr remisiones prolongadas, sino de   cura  de ciertos tipos de cancer,  con solo  copiar  y mejorar  las  defensas celulares normales  de nuestra propia humanidad  (Shawn Hingtgen: Tumor-homing cytotoxic human induced neural stem cells for cancer therapy.Translational Medicine. Vol. 9 Issue 375).   Un caso mas del correcto uso de  la  medicina natural. 1)  El articulo relata observaciones realizadas en ratones y  humanos con tumores cerebrales (glioblastomas), en las que celulas madre neurales (NSCs, neural stem cell),   reprogramadas a partir de celulas autologas adultas de piel, migran   hacia las celulas tumorales  en respuesta  a señales quimiotacticas (tumor homing properties),  liberadas por las celulas tumorales, infiltrando   focos primarios e invasivos de los glioblastomas,   reduciendo   el tamaño de las celulas tumorales al  2% o 5%,  de su tamaño original.  Una novedad terapeutica,  ya que la cirugia,  quimioterapia o radiacion son incapaces de erradicar  las recurrencias de glioblastomas. 2) Hay que considerar que   es dificil  obtener  NCSs, autologas  y que las NSCs,  procedentes de donadores tienen el riesgo de rechazo y otros eventos colaterales.  Bagó J.R., obvio  este riesgo empleando fibroblastos de piel de ratones  a las que  transdiferencio a  NSCs,  un proceso  que en ratones dura  4 dias. Obtenidas,  las NSCs humanas,   son inyectadas directamente en los glioblastomas  cerebrales reduciendo   20 a 50 veces su tamaño, en 24-28 dias. En humanos, las  NSCs, aparte de  infiltrar a las  celulas tumorales,    liberan en  los glioblastomas :  terapia citotoxica,    productos geneticos, sustancias proapoptoticas, factor alfa de necrosis tisular, etc.,   reduciendo  250 veces  el tamaño de los glioblastomas   en 3 semanas (Methods. 2016.Neural stem cell therapy for cancer). ADOPTION OF GROUP BEHAVIOUR



HOW BIO-MACHINES MAKE DECISIONS

Jessica Flack, an evolutionary biologist who works at Saint Fe Institute (USA), has been doing studies for two decades to understand the use of computer processes by parts of organs or groups of organisms (neural networks, bird  flocks, eukaryotic cell clusters with reproductive purposes, etc.), to adopt group behaviors and solve daily adaptive problems. After scrupulously analyzing   decision-making processes in primates, Flack   tries  now  to extend these studies to neuronal collectives of animals and humans, of course. Interestingly, an experiment conducted at Stanford by Bill Newsome and coworkers on macaques who were going to decide whether a group of points on a screen would move to the right or left. The conclusions of the study were: 1) At the beginning of the task, few   independent  macaque neurons processed the data to make a decision. 2) However, as the definitive moment approached, the pattern changed and similarities were observed in the activity of the neurons in order to agree to make the chosen decision, finally starting a collective computation process chosen from the noisy input, unifying the activities, Establishing a consensus about which decision to take (principle of accumulation of information), applied by certain societies of monkeys to win battles. Subsequently, the information was strengthened through an exchange of special signals. Decoding of these, allows to understand how much consensus existed in the group, regarding the ability of an individual (s), to use force in their battles. Flack attempts to explore the possibility that few formidable fighters  could  stabilize a whole group of monkeys  intimidating weak monkeys showing   them teeth rather than risking a fight they might lose. Or, eliminate the police group from the opposing side, leading it to chaos. Flack tries to understand how microscopical phenomenological rules can   reach beneficial macroscopic behaviors, to understand how groups solve their adaptive problems and make decisions, why  complex systems are held together by small circles of power. Focused on information, Flack tries to copy the processes that will lead us to make correct decisions, associating them with emerging learning machines in order to solve political, market and other problems. A further argument in favor of our theory that humans are biomachines.

COMO TOMAN DECISIONES LAS BIOMAQUINAS

Jessica Flack,  bióloga evolutiva que  labora   en  el Instituto  Santa Fe (USA),  realiza estudios desde hace 2 décadas,  para  comprender  el empleo de  procesos informáticos por  partes órganos o grupos de organismos (redes  neuronales,  bandadas de palomas, agrupaciones de células eucariotas con fines reproductivos, etc.), para adoptar conductas grupales  y  resolver  problemas adaptativos cotidianos.  Tras analizar escrupulosamente  los procesos de toma   de  decisiones  en macacos (primates), Flack intenta   ahora, extender   estos estudios a colectivos neuronales de animales  y  de  humanos, por supuesto. Resulta interesante  al respecto, un experimento realizado en Stanford por   Bill Newsome y colaboradores en   macacos que iban a   decidir si un grupo de puntos insertos en una pantalla se movería a  la derecha o a la  izquierda. Las conclusiones del estudio  fueron: 1) Al inicio de la tarea,  pocas neuronas independientes  de los macacos procesaron  los datos para tomar una decisión. 2) Empero, conforme se aproximaba el momento definitivo, el patrón cambiaba  notándose similitudes en la actividad de las neuronas a fin  de  ponerse de   acuerdo  para tomar la decisión  elegida,  iniciándose finalmente un proceso de computación colectiva escogida  del  noisy input, unificándose las  actividades,  estableciéndose un  consenso acerca de que decisión tomar (principio de acumulación de información),  aplicado por ciertas  sociedades de macacos  para  ganar batallas. Posteriormente, la información  se afianzo mediante un intercambio de señales  especiales.  La  decodificación de estas, permite  entender   cuanto consenso existia en el grupo, respecto a la  capacidad de un individuo (s),  para usar la fuerza en sus batallas.  Aplicativamente,  Flack  intenta   explorar la posibilidad de que  pocos luchadores formidables   estabilicen   a un grupo entero de macacos  intimidando a   monos  débiles  enseñándoles los dientes más que arriesgándose a  una lucha que podrían perder.  O,  eliminar al grupo policial del bando contrario,   llevándolo al  caos.   Flack, trata de entender como a partir de reglas fenomenológicas microscópicas es posible arribar  a conductas macroscópicas beneficiosas, de entender  como los grupos resuelven sus problemas adaptativos y toman decisiones,  porque los  sistemas complejos se mantienen robustos unidos por pequeños círculos de  poder.  Focalizada en la información, Flack  intenta   copiar  los procesos que nos conducirán a tomar   decisiones   correctas, asociándolas a  emergentes   máquinas de aprendizaje a fin de  resolver  problemas políticos, de mercado  y  otros.    Un argumento más a favor de nuestra teoría de que los humanos somos  biomaquinas.
