SINGULATIES OF BLACK HOLES












BEFORE THE SINGULARITY, THE TIME EXISTED

When Georges Lemaitre observed 87 years ago, the displacement of light from distant galaxies, he understood that the universe was expanding, inducing him to postulate the theory of the cosmic egg (Big Bang), reinforced later by additional arguments of Gamow, Hubble, Penzias, etc., who said   that some 13.8 billion years ago, the initial universe was a state of infinite density (singularity) and the temperature was extremely high.  Soon after, the space-time expanded continuously, until today. When in September 2015, thanks to the Ligo experiment, the first direct observation of gravitational waves was made, which reinforced the concept of singularity, the Big Bang almost ceased to be a theory, leaving only to be explained what there was before the occurrence of this phenomenon. With what is currently known about black holes (the other side of the Big Bang, according to us), there are convergences between the Big Bang and black holes: both have singularities from which do not escape the light, nor the time. Thus, when we ask what happens with the singularity that results from the merger of multiple black holes in our universe, one option, depending on the greater or lesser amount of accumulated information, is that a gigantic black hole could originate a similar or a different new universe and so on, cyclically. It is known that when  two medium size black holes collide form  one of larger size. A small black hole could eventually originate a wormhole. Although well-informed scientists (S.Hawking),  argue that black holes lose information because of  inherently outward radiation, Hawking himself seems to contradict himself when, together with Penrose, claimed that quantum processes within singularities  are  incomplete and that they may need other types of mathematical equations. If the possibility of forming new universes from singularities formed in black holes would exist, the answer to an initial question would be that time existed before the Big Bang, provided by the preceding universe. In this regard, Tim Koslowski, Flavio Mercati and David (Physics Letters B), have reinterpreted the model of physical changes in singularities, when considering separately the proper  space-time from  the matter that it contains, replacing the singularity with a  Janus Point :  relative scales of the material that makes up the universe are crushed to set a 2D plane, as time is rewound. After that, the 2D plane (Janus point) recovers its 3D, although in the opposite direction, reaching deep implications in the symmetry of the physics of the possible particles, obtaining a universe based essentially on antimatter.



ANTES DE LA SINGULARIDAD, EXISTIÓ EL TIEMPO

Cuando   Georges Lemaitre observo hace 87 años, el desplazamiento de la luz procedente de  galaxias distantes, comprendido   que el universo se estaba expandiendo, induciéndolo  a postular  la teoría del huevo cósmico (Big Bang), reforzada más tarde  por  argumentos adicionales de Gamow, Hubble, Penzias,etc., quienes aportarían que hace unos 13 800  millones de años, el universo inicial era un estado de  densidad infinita (singularidad) y temperatura extremadamente altas, para poco después, expandirse en el tiempo-espacio en forma continua, hasta hoy.  Cuando en septiembre del 2015, merced al experimento Ligo, se realizó la primera observación directa de ondas gravitacionales, que reforzaron el   concepto de singularidad, el Big Bang casi dejo de ser una teoría, quedando solo por explicar que hubo antes de la ocurrencia de este fenómeno. Con lo que se sabe actualmente de los agujeros negros (la otra cara del Big Bang, según nosotros), se observan convergencias entre el Big Bang y los agujeros negros: ambos poseen en mayor o menor grado singularidades, de las que no escapan la luz, ni el tiempo. Así, cuando nos preguntamos que pasara con la singularidad que resulte de la fusión de múltiples agujeros negros en nuestro universo, una opción, dependiendo de la mayor o menor cantidad de información acumulada, es que   un gigantesco agujero negro origine un universo nuevo semejante o diferente al nuestro y así sucesiva y cíclicamente. Es conocido que agujeros negros de tamaño mediano colisionan formando uno de mayor tamaño.  Un agujero pequeño podría originar eventualmente, un agujero de gusano. Aunque científicos bien informados (S.Hawking), arguyen que los agujeros negros pierden información a causa de una radiación hacia afuera inherente a estos monstruos, el mismo Hawking parece contradecirse cuando junto con Penrose,  afirmaron  que los procesos cuánticos al interior de las singularidades al ser incompletas, podrían necesitar de otro tipo de ecuaciones matemáticas. Si la posibilidad de formar nuevos universos a partir de singularidades formadas en agujeros negros existiese, la respuesta a una pregunta inicial sería que el   tiempo existió antes del Big Bang, proveído por el universo precedente.  Al respecto, Tim Koslowski, Flavio Mercati y David (Physics Letters B), han reinterpretado el modelo de cambios físicos en singularidades, al considerar de manera separada al espacio-tiempo propiamente dicho, de la materia que contiene, remplazando la singularidad por el Punto de Jano: las escalas relativas del material que constituye el universo son aplastadas hasta configurar un plano de 2D,  a medida que se  rebobina el tiempo. Tras ello, el plano de 2D (punto de Jano), recupera sus 3D, aunque en sentido inverso, alcanzándose profundas implicaciones en la simetría de la física de las partículas posibles, obteniéndose un universo basado esencialmente de antimateria.

HELICAL SUN SYSTEM MOVEMENT









The helical movement of the solar system

The Sun has two movements: one of rotation on its own axis and one of translation around the Milky Way (our galaxy: about 100,000 light years in diameter), together with a giant cluster of stars. The sun translation movement is different from that carried out by the planets around the sun. To begin, we will say that the equatorial rotation time of the sun lasts 26 days and that of its poles :36. The solar system revolves around the nucleus of the Milky Way at a speed of about 828,000 km/h, using 226 million light years in making a total return. During the movement of translation around the Milky Way, the sun advances by helicoidal turns (as a corkscrew does): up and down. During this process, the planets of the solar system also rotate helically around the sun.  If we could visualize the translational movement from outside the Milky Way, we would perceive that the sun advances like a comet with its planets revolving around it. The solar system moves upwards, with respect to the plane of the Milky Way, with an inclination of 63 °, angle best appreciated during a clear night sky.The sun and the celestial bodies that accompany it travel around the Milky Way in a wave form, moving from top to bottom and vice versa, for long periods. Frank Bash (University of Texas, Austin, Department of Astronomy), has calculated that within: 14 million years, the sun will reach its maximum translational height: 250 light years, above the galactic disk, then descend, reach the plane of the galaxy and then reach the point of 250 light years below the plane of the galactic disk and then ascend and so on. Like the rest of the celestial bodies of the Milky Way, the solar system is directed towards the center of the galaxy: the black hole of Sagittarius. Currently the sun is heading towards the star Lambda Hercules, at 72,000 km/h, on the way to becoming a white dwarf star, before being devoured by the big black hole. I) Why will the Sun and its planets have this translational helical movement? We do not know, but we intuit that this phenomenon happens to move faster, avoiding the enormous pressures of the surrounding gravity. II) Why is it heading towards the center of the galaxy? Although we do not know either, we intuit in the light of the known that the supermassive black hole is a kind of giant recycler of everything contained (energy and elementary particles), that after circulating through the  bridge  of a wormhole, is expelled at the other end, forming a new big bang and so on.

El movimiento helicoidal del sistema solar

El   Sol, tiene     dos movimientos: uno de rotación sobre su propio eje y otro de traslación en torno a la Vía Láctea (nuestra galaxia, de unos 100,000 años luz de diámetro), de conjunto con un cumulo gigante de estrellas. El movimiento de traslación solar es diferente al que realizan los planetas en torno al sol. Para empezar, diremos que el tiempo de rotación ecuatorial del sol dura 26 días y el de sus polos 36.    El Sistema solar gira alrededor del núcleo de la Vía Láctea a una velocidad de unos 828 000 km/h, empleando 226 millones de años luz en realizar una vuelta total. Durante el movimiento de translación alrededor de la Vía Láctea, el sol avanza mediante giros helicoidales (como lo hace un sacacorchos):  arriba y abajo. Durante este proceso, los planetas del sistema solar   giran también helicoidalmente    alrededor del sol.
Si pudiésemos visualizar el movimiento traslacional desde fuera de la Vía Láctea, percibiríamos que el sol avanza como un cometa con sus   planetas girando en torno a él.  El   sistema solar avanza dirigiéndose hacia arriba, respecto al plano de la Vía Láctea, con una inclinación de 63°, ángulo mejor apreciado durante un cielo nocturno despejado.    El sol y los cuerpos celestes que lo acompañan viajan alrededor de la Vía Láctea en forma de onda, moviéndose de arriba abajo y viceversa, durante largos periodos. Frank Bash (University of Texas, Austin, Department of Astronomy), ha calculado que dentro de: 14 millones de años, el sol alcanzara su máxima altura traslacional: 250 años luz, por encima del disco galáctico, para luego descender, alcanzar el plano de la galaxia y luego alcanzar el punto de 250 años luz debajo del plano del disco galáctico para luego   ascender y así, sucesivamente.  Al igual que el resto de los cuerpos celestes de la Vía Láctea, el sistema solar se dirige hacia el centro de la galaxia: el agujero negro de Sagitario. Actualmente el sol se dirige hacia la estrella Lambda Hércules, a 72000km/h, camino a devenir en una estrella enana blanca, antes de ser   devorado por el gran agujero negro, I) ¿Porque tendrán el Sol y sus planetas este movimiento traslacional helicoidal? No lo sabemos, pero intuimos que este fenómeno sucede para avanzar más rápido, sorteando las enormes presiones de la gravedad circundante.  II) ¿Porque se dirige hacia el centro de la galaxia? Aunque tampoco lo sabemos, intuimos a la luz de lo conocido que el agujero negro supermasivo es   una especie de reciclador gigante de todo lo contenido (energía y partículas elementales), para tras circular a través de la extensión de un agujero de gusano, ser expulsado por el otro extremo, formando un nuevo big bang y, así sucesivamente.

WALKING AGAIN






IT'S NOT THE BRAIN, IT'S THE SPINAL CORD

Based on which  dogma, 3 people with spinal cord injury (more  than 4 years),  could recover their ability to walk, using regulated and personalized electrical stimuli (Nature, 10/31/2018): to the simple and elementary knowledge that  movements of the legs are regulated by motoneurons located in the spinal cord and not in the brain  -explaining at the   same time-  the failure of previous techniques that prioritized brain neural reconnection with the adjacent neural spine cord. Although in most human beings, there are connections between the brain and motor neurons of the spinal cord, the function of neural brain connections is not to allow a person to move their legs, but rather to act like a director of an orchestra allowing fineness and art to more sophisticated movements. That is to say that the basic of the movement of the legs resides finally, in the spinal cord. In the last 5 years, the popularizer of this explanation is the Italian neurologist Sergio Canavero, who uses it to explain the projected head transplants to donated bodies, disconnected from the brain by accidents of various kinds. Faced with criticism that the transplanted head would not work properly, because the body would not walk, Canavero says that the patient with a transplanted head, will walk, because this function depends on the motor neurons of the spinal cord, very sensitive to electrical stimulation. Returning to the successful experiment of Gregory Courtine (Ecole Polytechnique Fédérale de Lausanne,Switzerland), we will say that his technique of electrical stimulation started experimentally in rats begins to generate impressive rehabilitations in human beings. Courtine, carried out this experiment (electrical epidural stimulation), in 8 people with spinal cord injuries with partial or complete lower limb paralysis, using a stimulator placed in the abdomen connected to an electrode field located in the lumbar region where there are cells that control the muscles of the legs. After the implant, Courtine, et al. developed a map of each patient to know where and how to apply the stimuli generated in real time.  The transplanted device is controlled wirelessly, when it is simulated the extension of the hip and the flexion of ankles that human legs do, when walking, inducing the motor neurons to generate activity. After a week of rehabilitation, the patients began to walk with partial support or with a walker, until they recovered their voluntary movements in the legs, without that support. Although the rehabilitation depends on the severity of the injury, the 8 participants improved, some more than others. According to Courtine, an alternate, precise and temporary stimulation is better than a continuous stimulation, blocking at the same time, residual signals that travel from the legs to the brain. The team was   able to program a sequence of electrical pulses that stimulated the spinal cord at the correct time and location to facilitate movements. Injuries to the spinal cord sever the connections between the brain and the motoneurons of spine, creating sensory and motor deficits in areas below the injury, sometimes producing paralysis. To make his work more precise, Courtin first mapped areas of the spinal cord, engaged in each movement, required to walk: hip or ankle flexion or extension. The electrical stimulation does not produce movement by itself, the patients themselves do it. The electrical stimulation reinforces the spinal brain-spinal cord neural connections, because the cells that previously discharged as a whole will continue unloading together.

NO ES EL CEREBRO, ES LA MEDULA ESPINAL

¿En base a que dogma, 3 personas con lesión medular con mas de 4 años de antigüedad, pudieron recuperar  su capacidad de   caminar,  empleando estímulos  eléctricos regulados y personalizados (Nature,31/10/2018): al sencillo y elemental conocimiento de  que los movimientos de las piernas son reguladas   por motoneuronas ubicadas en la  medula espinal y no  en el  cerebro, explicando  de paso, el fracaso de técnicas anteriores que priorizaban  la reconexión neural cerebral con la colindante neural  medular. Aunque en la mayoría de los seres humanos, existen conexiones entre el cerebro y las neuronas motoras de la medula espinal, la función de las conexiones neurales cerebrales no es permitir a una persona que mueva sus piernas, sino que al modo de un director de orquesta permitir fineza y arte a los movimientos más sofisticados. Es decir que la base del movimiento de las piernas reside finalmente, en la medula espinal.  En los últimos 5 años, el popularizador de esta explicación es el   neurólogo italiano Sergio Canavero, quien la emplea para explicar los proyectados trasplantes de cabeza a cuerpos vegetales donados, desconectados del cerebro por accidentes de diverso tipo. Ante las críticas de que la cabeza trasplantada no funcionaría adecuadamente, porque el cuerpo no caminaría, Canavero asegura, que   el paciente con una cabeza trasplantada, si caminaría, porque esta función depende de las motoneuronas de la médula espinal, muy sensibles a la estimulación eléctrica. Retornando al experimento exitoso de Gregorio Courtine (Instituto Politécnico de Lausanne, Suiza), diremos que su técnica de estimulación eléctrica iniciada experimentalmente en ratas empieza a generar   rehabilitaciones impresionantes en seres humanos. Courtine, llevo a cabo este experimento (estimulación epidural eléctrica), en 8 personas con lesiones medulares con parálisis parcial o completa de extremidades inferiores, empleando un estimulador colocado en el abdomen conectado a un campo de electrodos situado en la región lumbar donde se encuentran las células que controlan los músculos de las piernas.  Tras el implante, Courtine y colaboradores, elaboraron un mapa de cada paciente para saber dónde y cómo aplicar los estímulos generados en tiempo real.
El dispositivo trasplantado, se controla de forma inalámbrica, cuando se simula la extensión de la cadera y la flexión que hacen las piernas humanas al caminar, induciendo a las motoneuronas a volver a generar actividad.
Después de una semana de rehabilitación, los pacientes empezaron a caminar con apoyo parcial o con un andador, hasta recuperar sus movimientos voluntarios en las piernas, sin ese apoyo. Aunque la rehabilitación depende de la gravedad de la lesión, los 8 participantes mejoraron, unos más que otros.  Según Courtine, una estimulación alternada, precisa y temporal, es mejor que una   estimulación continua, bloqueando de paso, señales residuales que viajan de las piernas al cerebro. El equipo debe ser capaz de programar una secuencia de pulsos eléctricos que estimulen a la médula espinal en el tiempo correcto y localización para facilitar los movimientos. Las injurias a la médula espinal cortan las conexiones entre el cerebro y las neuronas de la médula creando déficits sensoriales y motores en áreas debajo de la injuria, induciendo a veces parálisis.  Para hacer su trabajo más preciso, Courtin primero mapeo áreas de la medula espinal, comprometidas en cada movimiento, requerido para caminar: flexión o extensión de cadera o tobillo. La estimulación eléctrica no produce movimiento por sí mismo, los propios pacientes lo hacen. La estimulación eléctrica refuerza las conexiones neurales cerebro-médula espinal, porque las células que antes descargaron de conjunto continuaran descargando conjuntamente.

GENOME EDITION





A SMALL STEP, A GREAT LEAP FOR HUMANITY

According to the chemist David Liu (MIT, Harvard University), a scientific revolution is brewing in the areas of chemistry-genetics-biology and immunology, of gigantic proportions. Although there are obviously emerging ethical problems, the techniques of editing the genome of living beings on planet Earth, begin to cure or alleviate a growing number of diseases. The transcendence of these techniques extends to the possibility of creating genetically improved human beings and extend the useful lifespan of humans, animals and plants. I) Liu, emphasizes that the process of replacement of defective nucleotides in live mice occurs in differentiated cells that no longer divide, generating cascades of molecular signals capable of self-repairing parts of the cochlea of ​​the inner ear, an important fact because half of the cases of deafness have genetic bases. Liu, who is aware of the emerging collateral effects of these techniques in the rest of the genome, argues that, by the way, nobody knows the side effects before developing a therapeutic application. However, when a disease is fatal, the ethical risks are minor. The scientist believes that the synergistic use of several genome editing strategies will reduce these risks. II) He does not believe in the emergence of immune reactions to this technique because many of these proteins of  CRISPR-Cas9, exist in the bacteria that inhabit our body. Aware of the rumors that these techniques could be used as weapons of mass destruction, Liu points out that there are more effective weapons for such purposes and that clinical trials are conducted in humans’ beings, not in petris. He adds that we must respect ethics, but we must not be so rigid, saying that ethical moratoriums cannot be global, although he agrees with some, for example, he is in favor of a moratorium banning experiments on human germ cells. He also believes that  genome  editing  (simple, selective and effective), will establish new therapies for diseases that depend on a specific mutation, improve techniques in agronomy through corrective genes and others. III) The scientist emphasizes that the CRISPR-Cas9 technique, belongs to genetic engineering, is inexpensive, uses a bacterial enzyme (Cas9), fragments of RNA and manipulates specific sequences, being applicable to rodent plants, monkeys, and human embryos, allowing introduce artificial mutations, in cells of culture, or in guinea pigs suspected of causing certain diseases, permit  study the effects and develop treatments, in mutations that cause: blindness, deafness, epilepsy, neurological or metabolic diseases and in cancers, stop their proliferation, allowing T lymphocytes, rearranged  with   genetic engineering  to attack  cancer cells. Of 64,000 genetic variants, associated with diseases, 2/3, are point mutations. IV) However, the initial CRISPR techniques, equivalent to molecular scissors, have certain problems, because they cut the DNA in a specific site in order to remove the unwanted gene, without recognizing that the cell itself is capable of joining by itself the damaged DNA parts.  Therefore, Liu and collaborators created the: Basic Editor, a protein that does not exist in its natural state, that has components extracted from nature, but also synthetic, that does not cut DNA, but that realizes a rearrangement of atoms, reconfiguring the defective nucleotides, a process aided by intracellular cameras, which visualize the activities of a viral infection, triggering editing mechanisms in the genome of the cells involved, allowing to read the historical sequence of reactions initiated by the cell. 2  others systems: Detector and Sherlock, detect the anomalous presence of foreign DNA, being helpful to determine the  characteristics of plant’s resistance, doing the same as farmers of a few decades ago when they crossed varieties of the same plant to transfer genes and improve them naturally.  Liu hopes that plants treated in this way will not be considered nor regulated as genetically modified organisms.

UN PEQUEÑO PASO, UN GRAN SALTO PARA LA HUMANIDAD

Según el químico David Liu (MIT, Harvard University), se está gestando   una revolución científica en las áreas de química-genética-biología e inmunología, de proporciones gigantescas.  Aunque obviamente existen problemas éticos emergentes, las   técnicas de edición del genoma de seres vivos en el planeta Tierra, empiezan a curar o paliar un número creciente de enfermedades. La trascendencia de estas técnicas se extiende a la posibilidad de crear seres humanos mejorados genéticamente y extender la vida útil de humanos, animales y vegetales.  I) Liu, enfatiza que el proceso de recambio de   nucleótidos defectuosos en ratones vivos se efectiviza   en células diferenciadas que ya no se dividen, generando cascadas de señales moleculares   capaces de autoreparar partes de la   cóclea del oído interno, hecho importante porque la mitad de los casos de sordera tienen   bases genéticas.     Liu, que es consciente de los efectos colaterales emergentes de estas técnicas en el resto del genoma, arguye que, por cierto, nadie conoce los efectos colaterales antes de desarrollar una aplicación terapéutica. Empero, cuando una enfermedad es fatal, los riesgos éticos, son menores. El científico, cree que el empleo sinergistico de varias estrategias de edición del genoma, reducirán estos riesgos. II) No cree en la emergencia de reacciones inmunes a esta técnica porque   muchas de estas proteínas (CRISPR-Cas9), existen en las bacterias que habitan en nuestro soma.     Enterado de los rumores de que estas técnicas podrían ser empleadas como armas de destrucción masiva, Liu retruca que existen armas más efectivas para tales propósitos y que los ensayos clínicos se realizan en humanos, no en petris. Agrega que hay que respetar la ética, pero no hay que ser tan rígidos, opinando que las moratorias éticas no pueden ser globales, aunque está de acuerdo con algunas, por ejemplo, está a favor de una moratoria que prohíba experimentos en   células germinales humanas.   Opina además que la edición del genoma (simple, selectiva y efectiva), establecerá nuevas terapias para  enfermedades que dependen de una mutación específica, mejorar técnicas en agronomía, mediante genes  correctores y otros. III)  Enfatiza  que la técnica   CRISPR-Cas9, pertenece a la  ingeniería genética, es poco costosa,  usa una enzima bacteriana  (Cas9),  fragmentos de  RNA y    manipula  secuencias específicas, siendo aplicable a  plantas roedores, monos, y embriones humanos, al permitir   introducir   mutaciones artificiales, en células de cultivo, o en  guinea pigs sospechosos de  causar  determinadas  enfermedades,  estudiar efectos y desarrollar tratamientos,  en  mutaciones que causan  :ceguera, sordera,  epilepsia, enfermedades neurológicas  o metabólicas y en canceres, detener su proliferación, permitiendo que    linfocitos T, modificados con  ingeniería genética ataquen  células cancerosas. De 64,000 variantes genéticas, asociadas a enfermedades, 2/3, son mutaciones puntuales. IV) Empero, las técnicas   CRISPR iniciales, equivalentes a    tijeras moleculares, tienen ciertos problemas, porque   cortan el   DNA en un sitio específico a fin de remover el gene no deseado, sin   reconocer que la propia célula es capaz de unir las partes deterioradas del   DNA.  Por ello, Liu y colaboradores crearon el:   "Basic Editor”, una proteína, que no existe en estado natural, que   tiene componentes extraídos de la naturaleza, pero también sintéticos, que no corta el   DNA, pero que realiza un   rearreglo de los átomos, reconfigurando los   nucleótidos defectuosos. Un proceso ayudado por “cámara intracelulares”, que visualizan las actividades de una infección viral, desencadenando   mecanismos de edición en el    genoma de las células implicadas, permitiendo leer la secuencia histórica de las reacciones iniciadas por la célula.  Otros 2   sistemas:   Detector and Sherlock, detectan    la presencia anómala de DNA extraño, siendo de ayuda para determinar las características de resistencia de las plantas, haciendo lo mismo que los agricultores de hace pocas décadas cuando cruzaban variedades de la misma planta para transferir genes y mejorarlos de modo natural. Liu espera que    las plantas tratadas de este modo no sean consideradas ni reguladas como organismos genéticamente modificados.

The Nobel Prize in Chemistry, 2018, was awarded to Frances H. Arnold (62, Caltech, Pasadena, USA), for controlling the directed evolution of certain enzymes, used to produce biofuels, improved medicines, etc. A triumph beyond the routine production of new materials and chemicals or rewriting the code of life. Triumph of perseverance, humility and recognition that the constructs of nature (not necessarily logical), are superior to our mental elaborations. According to part confession and after many frustrations for not being able to modify the enzymatic functions, FH Arnold, recognized the limits of the human mind in front of the enzymatic complexity (3D structure, constant mutations), adding that using only logic, contemporary thought and the more sophisticated computers, the task continued being difficult. She decided then, to adapt its plans to the canons established by the nature: reproduction, mutation, evolution. "I copied   the designs of nature." Initially, the scientist tried to change the properties of the enzyme subtilisin E, so that, instead of catalyzing reactions in aqueous solutions, to see if it   worked in organic solvents (dimethylformamide: DMF). To do this: 1-She introduced random mutations in the DNA of the enzyme to be modified. 2-Introduced the mutations in bacteria, producing thousands of variants of Subtilisin E, in order to establish which of them worked better in the organic solvent, a process that in directed evolution is known as selection. 3-When finding a variant (with 10 mutations), subtilisin E, which worked 256 times better in DMF, than the original enzyme, she created the directed evolution of the enzymes, producing more ecological enzymes for the pharmaceutical industry. Smith used bacteriophages (viruses that infect bacteria) to clone genes, a complicated task for the time. 1-He introduced a gene in the DNA of the protein capsule of a phage, reinserting the genetic mixture into a bacterium that produced a mass composed of millions of phages. 2-Smith then used the phage mass to find unknown genes of known proteins. His idea was to unite unknown genetic fragments, insert them into the phage DNA and then wait for the peptides to emerge from the surface of the phage. 3-When new phages are produced, phage proteins (containing the unknown gene), would appear on the surface of the phage forming part of the protein capsule. 4-Smith usually captured the phage using an antibody determined to adhere to the peptide, capturing the peptide gene (phage display). Simple and bright idea, because antibodies can identify and bind to a simple protein hidden within thousands with great precision: The functions of phage are to be a link   between the known protein and its unknown gene. In 1990, one of the people who adopted the phage display technique was Gregory Winter, who wanted   to design antibodies to block various disease processes. Given the difficulties in using antibodies obtained from mice, Winter investigated the potential of the method: phage display, inserting on the surface of the phage, antibodies in the form of Y; waiting for foreign substances to adhere to one of the ends of these arms. Winter inserted genetic information in this part of the antibody and DNA in the proteins of the phage capsule. In 1990, he   showed that an antibody determined to adhere to a small molecule (phOx) of the phage was sufficient to bind the phage with the antibody by extracting it from a mass of 4 million phages. Later, Winter showed that the phage display was useful in the directed evolution of antibodies. He  built a library with millions of varieties of antibodies on their surfaces, choosing from there those that adhere to different protein targets. Then he randomly changed the first generation of antibodies creating a new library where he found antibodies with even stronger adhesions to the target. In 1990  he created the human antibody: adalimumab, which neutralizes the protein: TNF-alpha, promoter of inflammation in many autoimmune diseases, including rheumatoid arthritis, psoriasis and intestinal inflammatory diseases. The phage display method has also been used to produce anti-cancer antibodies. A reflexive review of the works of the Nobel Prizes of Medicine, Physics and Chemistry, 2018, provides an extremely favorable balance for world science and technology. A merit to the talent of the authors of the papers, but most of all to the increasingly accelerated techno-scientific advances -a true breakthrough - thanks to the advances in information sciences, the reduction of the size of chips, nanotechnology, the revolution of imaging and microscopy. Although something is missing: a revolution in world ethical practice ... Hopefully it will happen.


PREMIO NOBEL DE QUÍMICA, 2018

El premio Nobel de Química, 2018, fue otorgado a Frances H. Arnold (62, Caltech, Pasadena, USA), por controlar la evolución dirigida de ciertas enzimas, usadas para producir biocombustibles, medicamentos mejorados, etc. Un triunfo   más allá de la producción   rutinaria de   nuevos materiales y químicos o reescribir el código de la vida.  Triunfo de la perseverancia, la humildad y el reconocimiento de   que los constructos de la naturaleza (no necesariamente lógicos), son superiores a nuestras elaboraciones mentales.   Según confesión de parte   y tras  muchas frustraciones por no poder  modificar las funciones enzimáticas, FH Arnold, reconoció los límites de la mente humana frente a la complejidad enzimática (estructura 3D, constantes mutaciones), añadiendo  que empleando solamente la lógica, el pensamiento contemporáneo y las computadoras más sofisticadas, la tarea continuaba siendo  difícil,   Se decidió  entonces, a  adecuar sus planes a los cánones establecidos por la naturaleza: reproducción, mutación, evolución. “Copie los diseños de la naturaleza”.  Inicialmente, la científica intento cambiar las propiedades de la enzima subtilisin E, para que, en vez de catalizar reacciones en soluciones acuosas, trabajase en solventes orgánicos (dimethylformamide: DMF).  Para ello :1-Introdujo mutaciones al azar en el DNA de la enzima por modificar. 2-Introdujo las mutaciones en bacterias, produciendo miles de variantes de la Subtilisin E, a fin de establecer   cuál de ellas trabajaba mejor, en el solvente orgánico, un proceso que en la evolución dirigida es conocida como    selección. 3-Al encontrar   una variante (con 10 mutaciones), de la subtilisin E, que trabajaba 256 veces mejor en DMF, que la enzima original, creo la evolución dirigida de las enzimas, produciendo enzimas más ecológicas para la industria farmacéutica. Ahora existen enzimas que transforman azucares simples en isobutanol.  El Premio Nobel fue compartido con George P. Smith (University of Missouri, Columbia, USA) y Sir Gregory P. Winter (MRC Laboratory of Molecular Biology, Cambridge, UK), por la “presentación de péptidos y anticuerpos en la superficie de fagos(phage display of peptides and antibodies).  En 1980, G. Idea simple y brillante, porque los anticuerpos pueden identificar y unirse a una proteína simple escondida dentro de miles con gran precisión: Las funciones del fago son ser un puente   entre la proteína conocida y su gen desconocido. En 1990, una de las personas que adopto la técnica del phage display fue Gregory Winter, quien anhelaba diseñar anticuerpos para bloquear varios procesos de la enfermedad. Dadas las dificultades para emplear anticuerpos obtenidos de ratones, Winter investigo el potencial del método: phage display, insertando en la superficie de los fagos, anticuerpos en forma de Y; esperando que en uno de los extremos de estos brazos se adhirieran sustancias foráneas, extrañas.   Winter inserto   información genética en esta parte del anticuerpo y DNA en las proteínas de la capsula del fago.  En 1990, demostró que un   anticuerpo determinado a adherirse a una pequeña molécula (phOx), del fago bastaba para unir al fago con el anticuerpo extrayéndolo de una masa de 4 millones de fagos. Después, Winter demostró que el phage display era útil en la evolución dirigida de anticuerpos. Construyo una biblioteca con millones de variedades de anticuerpos en sus superficies, escogiendo de allí a los que se adherían a diferentes objetivos proteicos.  Después al azar cambio la primera generación de anticuerpos creando una nueva librería en donde encontró anticuerpos con adherencias aún más fuertes al objetivo. En 1990 creo el anticuerpo humano: adalimumab, que neutraliza a la proteína:  TNF-alpha, promotor de la inflamación en muchas enfermedades autoinmunes, incluyendo la artritis reumatoide, psoriasis y enfermedades inflamatorias intestinales.  El método phage display, también ha sido empleado para producir anticuerpos anticancerígenos.  Un repaso reflexivo a los trabajos de los Premios Nobeles de Medicina, Física y Química, 2018, proporciona un saldo extremadamente favorable para la ciencia y la tecnología mundial. Un mérito al   talento de los autores de los papers, pero más que todo a los cada vez más acelerados avances tecnocientíficos -un verdadero breakthrough – a mérito de los adelantos en ciencias de la información, la reducción del tamaño de los chips, la nanotecnología, la revolución de la imagenología y la microscopia. Aunque falta algo: una revolución de la ética practica mundial…Ojalá se produzca.

NOBEL  OF PHYSICS, 2018

The Nobel Prize in Physics, 2018, was awarded this year to Arthur Ashkin (96, Cornell University, Ithaca, USA), for having invented optical laser light tweezers, capable of capturing very small particles, atoms, viruses, and others. For this, Ashkin used the central pressure of the laser light, to push physical objects moving them first towards the central part of the laser light where the intensity of the light was greater, keeping them there. But the most relevant work of Ashkin, was the one made in 1987, when after focusing 2 beams of laser light as optical tweezers, he captured live bacteria without damaging them. This type of clamp is now widely used to investigate the life machinery of various biological systems. The Prize was also received by Gérard Mourou (73, École Polytechnique, Université Paris-Saclay) and by Donna Strickland (58, University of Rochester, USA), for having taken the initial steps for the generation of the most intense laser pulses created by humanity up to now, published in 1985 in Strickland's doctoral thesis. Using an ingenious method, both were successful in creating ultrashort pulses of high intensity laser without destroying the amplified material. First, they amplified the laser pulses over time in order to reduce the power of their peaks, to finally compress them. If a pulse is compressed in time, becoming shorter and more light is packaged in the same small space, whereby the intensity of the light increases dramatically. In this way: Strickland and Mourou invented a new technique called: chirped pulse amplification (CPA), a standard for subsequent high intensity lasers. Next to millions of corrective eye surgeries performed every year using high precision laser beams, countless medical and industrial applications, are pending exploration.  An  extraordinary revolution in laser physics.

El Premio Nobel de Fisica,2018, fue otorgado este año a Arthur Ashkin (96, Cornell University, Ithaca, USA), por haber inventado   pinzas ópticas de luz laser, capaces de capturar partículas muy pequeñas, átomos, virus, y otros. Para ello, Ashkin empleo la presión central de la luz  laser, para  empujar  objetos físicos movilizándolos primero hacia la parte central de la luz laser donde la intensidad de la luz era mayor, manteniéndolos allí. Empero el trabajo más relevante de Ashkin, fue el realizado en 1987, cuando tras   focalizar 2 haces de luz laser a modo de pinzas ópticas, capturo bacterias vivas sin dañarlas. Este tipo de pinzas es ahora ampliamente utilizado para investigar la maquinaria de la vida de diversos sistemas biológicos.  El Premio, fue también recibido por Gérard Mourou (73, École Polytechnique, Université Paris-Saclay) y por Donna Strickland (58, University of Rochester, USA), por haber dado los pasos iniciales para la generación de los pulsos de laser más intensos creados por la humanidad hasta ahora, publicados en 1985 en la tesis doctoral de   Strickland. Empleando un método ingenioso, ambos tuvieron éxito en la creación de pulsos ultracortos de laser de alta intensidad sin destruir el material amplificado. Primero, amplificaron los pulsos laser en el tiempo a fin de reducir el poder de sus picos, para finalmente comprimirlos.  Si un pulso es comprimido en el tiempo, deviniendo   en más corto, se empaqueta más luz en el mismo pequeño espacio, con lo cual la intensidad de la luz se incrementa dramáticamente. De este modo: Strickland y Mourou inventaron una nueva técnica denominada: amplificación de pulsos gorjeada (CPA: chirped pulse amplification), un standard para subsecuentes láseres de alta intensidad. Al lado de millones de cirugías oculares correctivas ejecutadas cada año empleando   rayos láseres de alta precisión, innumerables aplicaciones médicas e industriales, están pendientes de ser exploradas: una extraordinaria revolución en la física de rayos láser.

NOBEL PRIZE FOR MEDICINE or PHYSIOLOGY, 2018
This time the Nobel Prize in Medicine or Physiology 2018 was awarded to the American James P. Allison (69, University of Texas, Anderson Cancer Center, Houston, Parker Institute for Cancer Immunotherapy) and to the Japanese Tasuku Honjo (75 years, Kyoto University), to discover anticancer therapies based on the acceleration of the anticancer activity of T cells, after releasing it from its natural immune brakes. Allison had studied for more than 3 decades the protein: CTLA-4, which slowed the anticancer activity of T cells. By conceiving, that eliminating this brake would favor the immune attack of T antitumor cells, Allison procreated seminal therapies based on brake removal of the activity of the immune system. Simultaneously, Tasuku Honjo discovered another protein in T cells, which also slowed down its activity, albeit with a different mechanism of action. In the last decade, the effectiveness of these therapies has been demonstrated, whether against metastatic melanoma, lung cancer, kidney, liver, lymphomas, etc. Conceptually, to the well-known strategies to fight against cancer: surgery, radiotherapy, chemotherapy, bone marrow transplantation, etc. another one is added with its own weight based on the control and manipulation of the immune system, useful even against advanced cancers, always more difficult to treat. Once again, the Nobel Prize in Medicine or Physiology is awarded to observations made by curious minds to that vast pharmacy and conglomerate of defensive systems generated in the human organism: a Nobel Prize awarded to researchers who copied defensive systems generated by the human soma. The breaking point of the human immune system is to discriminate the self   from non-self, being the bacteria, viruses, etc, recognized and eliminated, a system in which the T cells have a fundamental role. These cells have receptors that after joining foreign structures activate the immune defense system. In this cell area there are protein accelerators of the activity of T cells, along with other proteins that function as brakes of the activity of T cells, in which a balance prevents excessive activation that can lead to an immune destruction of cells and healthy tissues. After discovering in 1990 the protein: CTLA-4, Allison realizes that this functioned as a brake on the activity of T cells. In response, Allison generates an antibody against this protein, blocking its function. Eliminating the brake triggered the attack of immune cells against cancer. In 1994, Allison successfully treated mice with cancer, which were cured. Later humans with advanced melanoma would be cured. For its part, Tasuku Honjo would do the same using another protein-brake: PD-1, also expressed on the surface of T cells, although operating differently. The 2012 Honjo demonstrated the effectiveness of blocking PD-1 protein by achieving long remissions and the possible cure of metastatic melanoma. The new anti-cancer immune therapy has changed the prognosis of certain groups with advanced cancer. However, it develops certain serious adverse effects (immune hyperresponsiveness), usually manageable. At the moment the therapy against PD-1 is more effective with positive results against lung, kidney, lymphoma and melanoma cancer. Also, combination therapies (against CTLA-4 and PD-1) are more effective.

Esta vez el Premio Nobel en Medicina o Fisiología 2018, fue otorgado al americano James P. Allison (69 años, University of Texas, Anderson Cáncer Center, Houston, Parker Institute for Cancer Immunotherapy) y al japonés Tasuku Honjo (75 años, Kyoto University), por descubrir terapias anticancerígenas basadas en la aceleración de la actividad anticancerígena de las células T, tras liberarla de sus frenos naturales inmunes.  Allison había estudiado desde hace más de 3 décadas   a la proteína: CTLA-4, que frenaba la actividad anticancerígena de las células T.  Al concebir, que   eliminar este freno favorecería el ataque inmune de las células antitumorales, Allison procreo terapias seminales basadas en la eliminación de frenos de la actividad del sistema inmune.  Simultáneamente, Tasuku Honjo descubría otra proteína en las células T, que también frenaba su   actividad, aunque con un mecanismo de acción diferente. En la última década se ha demostrado la eficacia de estas terapias ya sea contra melanoma metastásico, cáncer de pulmón, riñón, hígado, linfomas, etc.  Conceptualmente y como se ve, a las ya conocidas estrategias de lucha contra el cáncer: cirugía, radioterapia, quimioterapia, trasplante de medula ósea, etc. se agrega otra con peso propio basada en el control y manipulación del sistema inmune, útil incluso contra canceres avanzados, siempre más difícil de tratar.   Una vez más se otorga el Premio Nobel de Medicina o Fisiología a observaciones realizadas por mentes curiosas a esa vasta farmacia y conglomerado de sistemas defensivos generados en el organismo humano:  un premio Nobel otorgado a investigadores que copiaron sistemas defensivos generados por el   soma humano.   El punto de quiebre del sistema inmune humano, es discriminar lo propio de lo extraño, siendo las bacterias, virus,etc,  reconocidos y eliminados, un sistema en el que las  células T tienen un rol fundamental.  Estas células tienen receptores que después de unirse a estructuras foráneas activan el sistema de defensa inmune.  En esta área celular existen   aceleradores proteicos de la actividad de las células T, al lado de otras proteínas que funcionan como frenos de la actividad de las células T, en el que un balance evita una excesiva activación que puede llevar a una destrucción inmune de células y tejidos saludables. Tras descubrir en 1990 a la proteína: CTLA-4, Allison se da cuenta que esta   funcionaba como un freno de la actividad de las células T. Ante ello, Allison, genera un   anticuerpo contra esta proteína, bloqueando su función. Al eliminar el   freno se desencadena el ataque de las células inmunes contra el cáncer.  En 1994 Allison trata con éxito a ratones con cáncer los mismos que se curaron. Después se curarían   humanos con  melanoma avanzado. Por su lado, Tasuku Honjo haría lo propio empleando otra proteína-freno:  la PD-1, también expresada en la superficie de las células T, aunque operando de modo diferente. El 2012 Honjo demostró la eficacia del bloqueo de la proteína PD-1 logrando largas remisiones y la posible cura del melanoma metastásico. La nueva terapia anticancerígena   inmune, ha cambiado el pronóstico de ciertos grupos con cáncer avanzado. No obstante, desarrolla ciertos efectos adversos serios (hiperrespuesta inmune), usualmente manejables.  De momento la terapia contra el PD-1 es más efectiva con resultados positivos contra cáncer de pulmón, renal, linfoma y melanoma. Asimismo, las terapias de combinación (contra   CTLA-4 y PD-1), son más efectivas.

A remarkable advance in the knowledge of the properties of   animals and human beings stem cells,  is the discovery of their rapid dedifferentiation towards a fetal state in response to severe tissue injuries in order to self-repair the affected body areas, an event known now as: paligenosis (return to a generative state), i.e.: cells that revert to a primitive state of rapid division using conserved  pathways  of  fetal deveploment. A better understanding of this process to know that a dedifferentiated stem cell, gives rise to another stem cell with normal differentiation capacity, while the other conditioned to self-tissue repair, develops a genetic program similar to that of a developing fetal organ. Knowledge that seen in perspective could promote the regeneration of whole members in humans’ beings, from structural buttons (rudiments of limbs), located in specific body sites. Process not exempt of certain risks because some dedifferentiations could not fulfill their self-repairing role, becoming rather in cancers (Florián R Greten and Sarah Schwitalla). Actually, some rechearchers are looking for markers of fetal programs useful to understand this process, which is known to allow wound healing in the intestines of the mouse. On the other hand, while investigating the mechanisms of immune reactions, Richard Locksley (immunologist) and Ophir Klein (stem cell biologist), both of the California SFC University, found that  in this process  certain responses were activated,  the Sca-1 gene expressed itself at high levels in the damaged tissue and that when culturing the proliferating cells of the injured area it was notorious that the Sca-1 genes formed fetal-like tissues, findings better understood when reviewing concepts of fetal bowel development, in molecular terms. Klein also noted that a series of genes expressed during fetal development had been transiently reactivated, with the same cellular reprogramming occurring after irradiations and inflammatory bowel injuries. Under such circumstances, the cells seemed to invoke a certain kind of fetal memory. Marie-Isabelle García (Université Libre de Bruxelles), was one of the first researchers to note the importance of the re-expression of fetal growth genes as part of a regenerative process. García, identified a protein marker in the progenitor cells of the stomachs of injured mice previously identified in progenitor cells of a fetal stomach and Kim Jensen (University of Copenhagen), identified in the colon of a mouse with inflamed intestines part of this process, explaining that the mechanical forces of the extracellular matrix that surrounded and sustained the activated cells carried a signal pathway that initiated tissue repair. The tissue matrix in this case became rigid, suggestive of a fetal state. Some scientists believe that this system of fetal tissue repair would have been left there, as a security system, which quickly builds its state, amplified the number of cells, not excluding the possibility of the development of a cancer that now most experts attribute to  being the final expression of a chronic inflammatory disease. Kim Jensen (University of Copenhagen) adds that the possibility of cancer increases when the human body faces physical trauma or when a chronic inflammatory bowel disease is installed. It has also been observed that the main cells of the stomach and the mature acinars of the pancreas with secretory functions in their tissues undergo fetal changes in response to injury, losing their natural adult markers, which are replaced by genes similar to embryonic ones, although lacking its characteristics of differentiation. The process of dedifferentiation towards a fetal state, is now known as: paligenosis: cells that revert to a primitive state of rapid division using conserved pathways of fetal development, whose objective is replication, rather than secretion. In this context, cancer would be the result of an erroneous paligenosis. Lately Jason Mills (University of Washington), has managed to identify some genes and compromised pathways having a perspective of connecting  findings tending to define a common mechanism capable of explaining regenerative processes and cancer, focusing now the study of these systems in their biology,  structural and signal mechanisms in each cell, capable of triggering similar responses to fetal ones, with the intention of suppressing function of mature, original stem cell.

AUTOCURACION, OTRA VEZ

Un notable avance en el conocimiento de las propiedades de las células madre adultas de animales y humanos es el descubrimiento de su rápida desdiferenciación hacia un estado fetal como respuesta a injurias tisulares severas a fin de auto-reparar las zonas   corporales afectadas, un evento conocido ahora como: paligenosis (retorno a un estado generativo) : células que revierten a un estado primitivo de rápida división empleando secuencias conservadas de las vías fetales. Un proceso mejor entendido al conocerse que una célula madre desdiferenciada, da origen a otra célula madre con capacidad de diferenciación normal, mientras la otra condicionada a la auto reparación tisular desarrolla un programa genético similar al de un órgano fetal en desarrollo.   Conocimiento que visto en perspectiva podría impulsar la regeneración total de miembros enteros en humanos, a partir de botones estructurales (rudimentos de miembros), ubicados en sitios corporales específicos.  Proceso no exento de ciertos riesgos     porque algunas desdiferenciaciones podrían no cumplir su rol autoreparador, deviniendo más bien en canceres (Florián R Greten and Sarah Schwitalla). Por lo pronto se buscan   marcadores de programas fetales útiles para entender   este proceso, del que se sabe que permite una cicatrización de heridas en los intestinos del ratón.   De otro lado y, mientras investigaba los mecanismos de las reacciones inmunes, Richard Locksley (inmunólogo) y Ophir Klein (biologo, especializado en células madre), ambos de la  Universidad   CSFC), encontró que ciertas respuestas   eran activadas, que el gene Sca-1 se  expresaba en altos niveles en el tejido dañado y que al cultivar las células proliferantes del área injuriada  era notorio  que los genes Sca-1, formaban   tejidos  semejantes a los fetales, hallazgos mejor comprendidos  al revisar  conceptos del desarrollo del intestino  fetal,  en términos moleculares.   Klein noto, asimismo, que   una serie de genes expresados durante el desarrollo fetal, habían sido transitoriamente reactivados, ocurriendo la misma reprogramación celular después de irradiaciones e injurias intestinales inflamatorias. En tales circunstancias, las células parecían invocar cierta clase de memoria fetal.    Marie-Isabelle García (Université Libre de Bruxelles), fue una de las primeras investigadoras en notar la importancia de la re-expresion de genes de crecimiento fetal a la que atribuyeron funciones regenerativas. García, identifico un marcador proteico en las células progenitoras de estómagos de ratones injuriados previamente identificados en células progenitoras de un estomago fetal. Kim Jensen (University of Copenhagen), identifico en el colon de un ratón con intestinos inflamados, parte de este proceso, explicando que las fuerzas mecánicas de la matriz extracelular celular que rodeaban y sostenían a las células activadas portaban una vía de señales que iniciaban la reparación tisular. La matriz tisular en este caso se tornaba   rígida, sugerente de un estado fetal. Algunos científicos creen que este sistema de   reparación tisular fetal habría sido dejado ahí, como un sistema de seguridad, que rápidamente construye su estado, amplificando el número de células, no exceptuándose la posibilidad del desarrollo de un cáncer al que ahora la mayoría de los expertos le atribuyen ser la expresión final de una enfermedad inflamatoria crónica.   Kim Jensen (University of Copenhagen), agrega que la posibilidad de un cáncer se incrementa cuando el organismo humano se enfrenta a traumas físicos o cuando una enfermedad intestinal inflamatoria crónica se encuentra instalada. Asimismo, se ha observado, que las células principales del estómago y las acinares maduras del páncreas con funciones secretorias en sus tejidos, sufren cambios fetales   en respuesta a la injuria perdiendo sus marcadores naturales adultos, los que son reemplazados por genes similares   a los embrionarios, aunque carentes de sus características de diferenciación. El proceso de desdiferenciación hacia un estado fetal, es conocido ahora como: paligenosis :  células que revierten  a un estado primitivo de rápida división empleando  secuencias conservadas de vías de desarrollo fetal, cuyo objetivo es la  replicación, más que la  secreción. En este contexto, el cáncer sería el resultado de una paligenosis errónea. Últimamente  Jason Mills (University of Washington), ha logrado identificar algunos genes y vías comprometidas  teniéndose ya una  perspectiva de hallazgos conectados tendientes a definir un mecanismo común capaz de explicar los procesos regenerativos y el cáncer,  focalizándose ahora el estudio de estos sistemas  en su biología celular,  mecanismos estructurales y de señales en cada célula,  capaces de   desencadenar respuestas similares a las fetales, con intenciones de  suprimir las funciones diferenciadas de   las células madre originales.
