INDIVIDUALIZED MEDICINE

One finishes inaugurating a new type of preventive medicine based on statistical probabilities and functions of specific genes. 2 days ago, Craig Venter and 30 scientists who work with him, announced to have decoded the first diploid genomic map (46 chromosomes), complete secuentiatión and assembly of 32 million fragments of human DNA, inherited of both parents, map which from now on (eliminating the selfish of some conservative scientists), must become the comparative standard of gold, of any human genomic sequence. Thanks to this map, the scientists specified that Venter has genes and loci, susceptible to certain diseases: alcoholism, coronary arteries disease, obesity, disease of Alzheimer, antisocial conduct and disorders of the conduct. The previous customized genome of Dr. James Wason, also revealed proclivity to the Alzheimer and certain type of cancer. With the aid of these genomic maps, it is hoped in the near future to make customized medicines quickly, without collateral effects and effective 100%. The new map eliminates the standard one completed the 2003, by the Human Genome Sequencing Consortium (academic centers financed by the American government), plenty of gaps (uncertain sequences of DNA). Although advanced, the map of the Consortium had some problems: it had sequenced only half (haploid), of the DNA (originated of people of different racial and ethnic origin), contained in a normal cell. On the contrary, the decoded genome of Venter, is complete (diploid), with DNA coming from both chromosomes (one of each father), pertaining to a single person: C. Venter.

The genome of Venter exhibits:I) 44% of heterozygous genes for one or but variant, that is to say: the copies of his mother differ from the inherited ones of his father. II) For this enormous task, Venter employed shotgun total genome sequencing (Sanger dideoxy method), that analyzes sequences of DNA of up to 800 units of length. The genomic version of Dr. Watson is being completed with a machine of last generation (454 Life Sciences), that orders the DNA pieces, by means of comparisons with the sequences of the Consortium’s genome. III) The genome of Venter, revealed the existence of 4500 gaps, 3 million SNPs: snips (small genetic change, or variation, that can occur within a person's DNA sequence, with consequences for the person), 1 million of indels (heterozygous events of insertion/deletion in a single unit of DNA), 4 million variations (single gene, existing in multiple copies), 90 inversions (a band of DNA, extracted and reinserted in reverse or mistaken site) and numerous segmental duplications IV) the genome of Venter, although confirms that a great number of genomic alterations, are located in single variations of nucleótides (SNPs: single nucleotide polymorphism), also revealed, that genomic variants little studied: (22% of insertions and deletions), give account of a 74% of variations, revealing that genetic variation between 2 individuals are 5-7 times but high (99.5%), that the considered one previously.

MEDICINA INDIVIDUALIZADA

Se acaba de inaugurar un nuevo tipo de medicina preventiva basado en probabilidades estadísticas y funciones de genes específicos. Hace 2 dias, Craig Venter y 30 cientificos que trabajan con él, anunciaron haber decodificado el primer mapa genómico diploide (46 cromosomas), completo (secuenciación y ensamblaje de 32 millones de fragmentos de DNA humano, heredado de ambos padres), mapa que a partir de hoy (eliminando el egoismo de algunos cientificos aferrados a concepciones antidiluvianas), debe convertirse en el standard de oro comparativo, de cualquier secuencia genómica humana. Merced a este mapa, los científicos explicitan que Venter tiene genes y loci, susceptibles a ciertas enfermedades : alcoholismo, enfermedad de arterias coronarias, obesidad, enfermedad de Alzheimer, conducta antisocial y desórdenes de la conducta. El genoma personalizado del Dr. James Wason, revelaba también proclividad al Alzheimer y cierto tipo de cáncer. Con la ayuda de estos mapas genómicos, se espera en un futuro cercano fabricar velozmente medicinas personalizadas, sin efectos colaterales y 100% eficaces. El nuevo mapa elimina al standard completado el 2003, por el Human Genome Sequencing Consortium (centros académicos financiados por el gobierno americano), lleno de gaps (secuencias inciertas de DNA). Aunque avanzado, el mapa del consorcio tenia algunos problemas : solo tenia secuenciada la mitad del DNA (proveniente de personas de diferente origen racial y étnico), contenido en una célula normal. Por el contrario, el genoma decodificado por Venter, es completo (diploide), con DNA procedente de ambos cromosomas (uno de cada padre), perteneciente a una sola persona : C. Venter.

I) El genoma de Venter exhibe: I) un 44 % de genes heterocigotos para una o mas variantes, es decir : las copias de su madre difieren de las heredadas de su padre. II) Para esta ciclópea tarea Venter empleó secuenciamientos shotgun totales del genoma (secuenciación de Sanger dideoxy), que analiza secuencias de DNA de hasta 800 unidades de largo. La versión genómica del Dr. James Watson se está completando con una máquina de ultima generación (454 Life Sciences), que ordena las piezas de DNA, mediante comparaciones con las secuencias del genoma del consorcio. III) El genoma de Venter, reveló la existencia de 4500 gaps, 3 millones de SNPs :snips (areas donde una sola unidad es cambiada, con consecuencias para la persona), 1 millon de indels (eventos heterocigotos de insercion/delecion en una sola unidad de DNA), 4 millones de variaciones (un solo gene, existiendo en multiples copias), 90 inversiones (una banda de DNA, extraida y reinsertada en un sitio equivocado) y numerosas duplicaciones segmentarias IV) El genoma de Venter, aunque confirma que un gran número de alteraciones genómicas, se localiza en variaciones de nucleótidos únicos (SNPs: single nucleotide polymorphism), reveló asimismo, que variantes genómicas poco estudiadas : (22% de inserciones y deleciones), dan cuenta de un 74% de variaciones de nucleótidos, revelando asi que la variación genética entre 2 individuos es 5-7, veces mas alta (99,5 %), que la estimada previamente.
