Primeros humanos alterados genéticamente.


En el experimento a ser realizado por cientificos de la Universidad de Newcastle, el núcleo de la célula A, (izquierda),con genes defectuosos mitocondriales, será extraído y trasplantado al huevo previamente enucleado, no fertilizado, de una mujer sana (célula B, derecha). De este modo los genes mitocondriales defectuosos, seran obviados.

Según Christian de Duve: http://nobelprize.org/medicine/laureates/1974/duve-autobio.html, (Premio Nobel de Medicina,1974), las primeras células (celulas mitocondriales, procariotas), eran cloroplastos o plástidos (estructuras que captan energía solar), envueltos en citoplasmas rudimentarios, nadando en variadas aguas. Sus genes en lugar de conformar un núcleo, solian estar dispersos en el citoplasma. La reproducción celular (clonación), se daba pese al discreto orden de sus genes dispersos. Más, al lado de estas, existían otras células, con mayor número de genes acondicionados al interior de un núcleo. En algún momento las células mitocondriales penetraron al interior de las células más grandes por un proceso de asimilación o, engulfamiento, a través de alguna grieta citoplásmica, dando como resultado células más eficientes (eucariotas), con 02 sets de genes: nucleares y mitocondriales. Es a partir de una de estas células, que se originaría el ser humano actual. De este modo al mismo tiempo que heredamos eficacia, también heredamos enfermedades (distrofias musculares, epilepsia, insuficiencia cardiaca, accidentes cerebrovasculares, demencia, tal véz infertilidad femenina), propias de genes mitocondriales defectuosos.

Con este conocimiento, el año 2001, varios investigadores del Instituto de Medicina Reproductiva y Ciencia de St. Barnabas, New Jersey, USA : http://news.bbc.co.uk/1/hi/sci/tech/1312708.stm, introdujeron DNA mitocondrial sano al interior del citoplasma (con DNA mitocondrial defectuoso), celular de un embrión humano temprano, proveniente de una mujer infértil. En este caso aunque el DNA mitocondrial defectuoso no fué removido, bastó la presencia del sano para contrarrestar los efectos nocivos del primero. Los reportes escritos de este experimento, señalan el nacimiento de 15 niños sanos. No obstante, en tanto la infertilidad, es aún causa discutible de afección mitocondrial, el experimento dejó de repetirse. Los criticos indicaron que el estudio era anético, tras comprobarse que uno de los niños sanos, portaba 3 sets genéticos : 01 de la madre, 01 del padre y 01 del donador.

Pero esta semana, cientificos de la Universidad de Newcastle (Inglaterra), consiguieron autorización de la máxima autoridad inglesa en fertilización y embriología (HFEA), no yá, para inyectar DNA mitocondrial sano, sino para implementar una variación técnica : http://news.bbc.co.uk/1/hi/health/4227320.stm. Ahora un núcleo embrionario humano incipiente, procedente de una persona con distrofia muscular genética, será extraido y transferido a un huevo sano enucleado, no fertilizado (ver: figuras y explicación arriba). De este modo, se espera que los descendientes carezcan de distrofia muscular (porque el DNA mitocondrial que las inducia será retenido en el embrión defectuoso, a ser descartado). Ahora las criticas vienen signadas por el hecho de que el huevo sano a ser fertilizado contendrá material génético de dos madres, lo que con toda seguridad alterará la línea germinal.
Se piensa que el experimento será exitoso, en tanto ya ha probado su factibilidad en ratones. De ser asi, el estudio abrirá las puertas a otros dos :1- aquel en el que el núcleo de un espermatozoide trasplantado a un óvulo femenino no fertilizado, hará las veces de huevo femenino. La posterior fertilización in vitro de este huevo, por espermatozoides de otro varón, hará posible lo inpensable: http://news.bbc.co.uk/1/hi/sci/tech/940790.stm, un niño, engendrado con el esperma de 2 varones homosexuales, y 2- la gestación de mujeres por mujeres (partenogénesis). Justo el dia de hoy, cientificos del Instituto Roslin (UK), http://news.bbc.co.uk/1/hi/sci/tech/4228992.stm, dan cuenta de los primeros embriones humanos logrados por estimulación eléctrica de óvulos femeninos sin fertilizar, sin intervención de espermatozoides masculinos.Estas dos posibilidades cientificas, las expliqué, hace 3 meses, en "Recreando a Eva del Edén". In the experiment to be made by scientifics of the University of Newcastle, the nucleus of one cell (left), with mitocondrials defective genes, will be extracted and transplanted to another egg previously enucleated, not fertilized, of a healthy woman (cell B, right). In this way, the defective mitocondrials genes, would be avoided.
According to Christian de Duve: http://nobelprize.org/medicine/laureates/1974/duve-autobio.html, (Nobel prize of Medicina, 1974), the first cells (mitocondrials cells, procariotes), were cloroplasts or plástides (structures that catches solar energy), surrounded in rudimentary cytoplasms, swimming in varied waters. Their genes instead of conforming a nucleus, were dispersed in the cytoplasm. The cellular reproduction (clonation), occurred in spite of the discreet order of its dispersed genes. More, next to these, other cells existed, with greater number of genes equipped to the interior of a nucleus. At some moment the mitocondrials cells penetrated to the interior of the greatest cells by an assimilation process or, engulfing, through some citoplasmic crack, giving more efficient cells (eucariotes), with 02 sets of genes: nuclear and mitocondrials. It is from one of these cells, that would originate the present human being. In this way we inherited effectiveness, and also inherited diseases (dystrophies muscular, epilepsy, cardiac failure, strokes, dementia, maybe female infertility), own of defective mitocondrials genes.
With this knowledge, in the year 2001, several investigators of the Reproductive Medicine Institute and Science of St. Barnabas, New Jersey, USA: http://news.bbc.co.uk/1/hi/sci/tech/1312708.stm, introduced healthy mitocondrial DNA to the interior of the cytoplasm (with defective mitocondrial DNA), cell of a human, early originating embryo of an infertile woman. In this case although defective, the mitocondrial DNA were not removed. The presence of healthy mitochondrial genes were enough to resist the injurious effects of the bad genes. The reports written of this experiment, indicate the birth of 15 healthy children. However, in as much the infertility, is still debatable as cause of mitocondrial affection, the experiment was not repeated. The critics indicated that the study was anéthic, after verifying themselves that one of the healthy children, carried 3 genetic sets : 01 of the mother, 01 of father and 01 of the donor.
But this week, scientifics of the University of Newcastle (England), obtained to authorization of the top English authority in fertilization and embryology (HFEA), not already, to inject healthy mitocondrial DNA, but to implement a technical variation: http://news.bbc..co.uk/1/hi/health/4227320.stm Now, an early human embryonic nucleus, coming from a person with muscular dystrophy, will be extracted and transferred to an enucleated healthy egg, not fertilized (see above: figures and explanation). In this way, one hopes that the descendants lack muscular dystrophy (because the mitocondrial DNA that caused this will be retained in the defective embryo, to being discarded). Now some criticize them signed by the fact that the healthy egg to being fertilized will contain genetic material from two mothers, whic maybe will alter the germinal line.
One thinks that the experiment will be successful, because it has already proven his feasibility in mice. If so, the study will open the doors to another two:1- one in which the nucleus of a spermatozoid transplanted to a non fertilized female ovum, will do the times of female egg. The later fertilization in vitro of this egg, by spermatozoa of another man, will make the impossible, possible: http://news.bbc.co.uk/1/hi/sci/tech/940790.stm, a boy, generated with the sperm of 2 homosexual men, and 2- the gestation of women by women (partenogénesis). Right the day of today, researchers of Roslin Institute (UK), http://news.bbc.co.uk/1/hi/sci/tech/4228992.stm, have been giving account of the first human embryos obtained by electrical feminine ovum stimulation without fertilizing, without intervention of male sperm. He escrito una Monografia:"La evolución",de modo que conozco el tema. Posiblemente los temas que trato y mis hipótesis son chocantes.Lo sé.Sin embargo,le ruego, revise la página de ciencia dela BBC del dia de ayer. Ahi comprobará lo que dije hace 3 meses:ya realizaron la primera partenogénesis humana.Tenga paciencia ya se realizarán mis otras hipótesis.Para comprender estas cosas hay que leer un poquito de biologia,genética,matemáticas y fisica. Como vé, poca cosa.
