CiPSCs CELLS



What is the meaning of obtaining chemically induced stem cells (CiPSCs), functionally similar to embryonic stem cells (ESC), using only a cocktail of small molecules to reprogram somatic cells to a pluripotent state able to generate tissues and organs of the body anywhere ?. I) CiPSCs cells, is a breaktrough that involves creating a simple, easy and safe way to induce a pluripotent state. II) Means not changing or adding genes (Oct-3/4, SOX2, c-Myc and Klf4) into cells to be reprogrammed, eliminating the risk of cancer and/or dangerous mutations. III) Means no use of retrovirus to transport transcription factors to DNA of somatic cells to be reprogrammed. IV) CiPSCs cells, sisters of induced stem cells (iPSC, created by Yamanaka), lack DNA methylation errors, maintain their ability to produce viable chimeras and apparently lack of limitations on next clinical applications . These cells are immunogenic, permeable, cheaper, easily synthesized, preserved, and easy to standardize to identify their effects to inhibit and activate specific protein function.

V) CiPSCs cells, also have potential therapeutic uses to remove the controversial use of human embryos. For the creator of these cells: Deng Hongkui (Peking University), this experiment involves the creation of a new route: inducing pluripotent state in somatic cell of immunodeficient mice by combining seven small molecule compounds, generating CiPSCs : 100% viable over 6 months, at a rate of 0.2%. Obtaining these 7 compounds demanded Deng, analyze 10,000 small molecules until finally find chemical substitutes to genes. Now it is possible to generate functionally desirable cell types in regenerative medicine, using specific chemicals or drugs instead of manipulating genes. Also to analyze the mechanisms of growth of new members, as one of the genes involved in cellular reprogramming (Sall4) is also involved in cellular dedifferentiation (reprogramming) of cells of amputees members of amphibians, before the growth or regrowth thereof.

¿Cual es el significado de la obtención de células madre químicamente inducidas (CiPSCs), funcionalmente similares, a células madre embrionarias empleando únicamente un coctel de pequeñas moléculas para reprogramar células somáticas a un estado pluripotencial capaz de generar tejidos y órganos en cualquier parte del cuerpo?. I) La creación de células CiPSCs, es un suceso tecnológico, que implica la creación de un método simple, fácil y seguro para inducir un estado pluripotencial. II) Significa no cambiar ni añadir genes (Oct-3/4, SOX2, c-Myc and Klf4), a las células por reprogramar, eliminando el riesgo de cáncer y/o de peligrosas mutaciones. III) Significa no emplear retrovirus para transportar los factores de transcripcción al DNA de las células somáticas por reprogramar. IV) Las CiPSCs, hermanas de las células madre inducidas (iPSC, creadas por Yamanaka), superan a estas últimas al carecer de los errores de metilación del DNA, mantener su capacidad para producir quimeras viables y al parecer carecer de limitaciones en futuras aplicaciones clínicas. No son inmunogénicas, son células permeables, mas baratas, fácilmente sintetizadas, preservadas, y fáciles de estandarizar para identificar sus efectos de inhibir y activar la función de proteínas especificas.

V) Asimismo, las células CiPSCs, tienen potenciales usos terapéuticos al eliminar el uso controversial de embriones humanos. Para el creador de estas células :Deng Hongkui (Universidad de Pekin), este experimento implica la creación de una nueva ruta, al inducirse un estado celular pluripotencial en células somáticas de ratones inmunodeficientes mediante la combinación de 7 compuestos de pequeñas moléculas, generando células CIPSCs 100% viables durante 6 meses, a una tasa de 0.2 %. La obtención de estos 7 compuestos le exigió a Deng, analizar 10,000 pequeñas moléculas hasta encontrar los sustitutos químicos a los genes. Ahora existe la posibilidad de generar tipos célulares funcionalmente deseables en medicina regenerativa, empleando químicos o medicamentos específicos en lugar de manipular genes. Asimismo permitirán analizar los mecanismos de crecimiento de nuevos miembros, ya que uno de los genes implicados en la reprogramación celular (Sall4), también participa en la desdiferenciación celular (reprogramación), de las células de los miembros amputados de los anfibios, antes del crecimiento o recrecimiento de los mismos.

ANOTHER STEP TOWARDS AIDS CURE

What does it means the negativization of serum, blood and tissue tests of 2 Boston/US patients previously HIV-positive with leukemia or lymphoma, after being subjected to bone marrow transplantation?. I) Such results contribute to design a cure for AIDS, as the absence of virus or antibodies to the virus in the blood and other tissues suggest the elimination of important viral reservoirs in the bone marrow, which should be a continuous generator of viral infected lymphocytes. Timothy Brown (42), was the first HIV-positive patient with acute leukemia, to receive a bone marrow transplant from a donor with an HIV resistant mutation. After transplantation, Timothy became seronegative for HIV. After 5 years without antiretroviral therapy, Timothy is now considered cured or in remission (the virus can lie dormant in the brain, gastrointestinal tract and other sites). Therefore urge new methods to detect and quantify HIV virus in hidden reservoirs II) Bone marrow transplantation has no practical use in HIV-positive people without cancer, since they live useful lives with alone antiretroviral therapy. Until we get enough knowledge about HIV virus, bone marrow transplants in HIV-positive patients should be performed only in patients with blood cancers. We need to add to the previous report the cure of a Mississippi/US, HIV-positive baby. Some researchers believe that antiretroviral drugs given at birth prevent the establishment of body viral reservoirs.
III) The reported cases encourage projects to create cells resistant to HIV infection. IV) There are some differences between Timothy and American patients with lymphoma: a) While the bone marrow of Timothy was completely destroyed with drugs and total body irradiation (mortality: 40%), the bone marrow of patients with lymphoma were only partially destroyed, a process that carries a 20% mortality. b) Timothy received a marrow from a close genetically related donor, with a rare mutation in the CCR5 receptor, a surface protein of CD4 cells which normally allows the HIV virus, enter, infect and destroy the immune system. CCR5 Delta 32 mutation causes the person who owns it to be resistant to HIV infection. In the future, genetic reengineering could artificially emulate CCR5 receptor mutation. c) Donors of Boston did not have the CCR5 Delta 32 mutation. d) Boston patients continued to receive antiretroviral therapy several years after transplantation. e) It is assumed that graft-versus-host disease generates cells that attack old infected tissues previously weakened by chemotherapy, removing it from the body: a process that lasts nine months. Although it can be fatal the graft-versus-host disease, could eliminate most viral reservoirs f) Sirolimus, seems to have helped to eliminate the HIV virus.

UN PASO MAS HACIA LA CURA DEL SIDA

¿Que implica la negativización de los tests sanguíneos, tisulares y serológicos de 2 pacientes de Boston-USA- previamente HIV positivos con leucemia o linfoma, tras ser sometidos a trasplantes de medula ósea?. I) Contribuyen a diseñar una cura para el SIDA, en tanto la ausencia de virus o anticuerpos al virus en la sangre y otros tejidos sugieren la eliminación de importantes reservorios virales en la medula ósea, la misma que debe ser un generador continuo de linfocitos infectados con virus. Timothy Brown de 42 años fue el primer paciente HIV positivo con leucemia aguda, en recibir un trasplante de medula ósea de un donador con una mutación resistente al H.I.V. Después del trasplante, Timothy devino en seronegativo al HIV. Tras 5 años sin recibir terapia antirretroviral Timothy es considerado actualmente curado o, en remisión (el virus puede permanecer dormido en el cerebro, tracto gastrointestinal y otros). Por ello, urgen nuevos métodos para detectar y cuantificar los virus HIV en reservorios ocultos II) El trasplante de medula ósea no tiene uso práctico en personas HIV positivos, sin cáncer, ya que estas viven vidas útiles con solo terapia antiretroviral. Mientras no se conozca los suficiente, los trasplantes de medula ósea en pacientes HIV positivos, deberían ser realizados solo en pacientes con canceres de la sangre. Es necesario agregar a lo anterior la cura de un bebe HIV positivo en Mississippi, USA. Algunos investigadores opinan que las drogas antiretrovirales administradas al nacer impiden el establecimiento de reservorios virales corporales.

III) Los casos reportados alientan proyectos para crear células resistentes a infección por HIV. IV) Se aprecian algunas diferencias entre Timothy y los pacientes americanos con linfoma : a) Mientras que la medula de Timothy fue completamente destruida con medicamentos e irradiación corporal total (mortalidad : 40%), la medula ósea de los pacientes con linfoma fueron solo parcialmente destruidas medicamente, un proceso que acarrea un 20% de mortalidad. b) La medula que recibió Timothy procedía de un donador genéticamente emparentado con una rara mutación en el receptor CCR5, una proteína superficial de las células CD4 que normalmente permite que los virus HIV, ingresen, infecten y destruyan el sistema immune. La mutación Delta 32 CCR5 hace que la persona que lo posea sea resistente a la infección por el virus HIV. En el futuro la reingeniería genética, podría emular artificialmente la mutación del receptor CCR5. c) Los donadores de Boston no tenían la mutación delta 32 CCR5. d) Los pacientes de Boston continuaron recibiendo terapia antiretroviral varios años después del trasplante. e) Se asume que la enfermedad graft-versus-host, genera células que atacan al tejido viejo infectado y debilitado por la quimioterapia eliminándolo del cuerpo: un proceso que dura 9 meses. Aunque puede ser fatal, la enfermedad graft-versus-host, eliminaría la mayoría de reservorios virales f) El Sirolimus, podria haber contribuido a eliminar al virus H.I.V.

PERU: 2 PREHISPANIC EMPIRES




I-According to Krzysztof Makowski (Univ. Católica del Perú: PUCP), burial remains found in the archaeological complex El Castillo (Huarmey/Ancash/Peru), are andean Wari style, by the characteristics of their burial chambers: plaster floors, walls that cross as a honeycomb, use of adobe (no stone), in its construction, for obvious reasons. Buried individuals are not extended as Moche, but sitting in Wari position with few lap wrappers. II-Why the discovery of 63 mummies (50 ladies of the nobility) and 1200 objects (silver earrings, gold necklaces, pectoral and good quality ceramic), found in the complex, would modify Peruvian prehispanic history and evolutive conception of civilizations in Peru and in the world?. Being located the complex El Castillo on the north coast of Peru, far from andean Wari (40 000 inhabitants, walled urban core), capital of Wari culture (600-1200 years a.c., 25 km NE of Ayacucho), solidifies the concept of Wari Empire. Makowski, agrees that Wari culture was an empire that exist before the Incas, given their way of conquering and spread. Wari culture rooted in Warpa tradition (300 years a.c.). First conceptions of pre-hispanic civilizations (Max Uhle), were made on the basis of archaeological macrodiscoveries : Chavin, Moche, Nazca, Tiahuanaco, Inca, saw as independent settlements, with limited communicating vessels between them.

III-Wari recent findings, confirm a centralized unit in prehispanic altiplanic andean with extensions to coast and jungle. Wari extended north to Piura, south to Cuzco, Arequipa and Tacna and east to the jungle (Lamas/San Martin). Julio C. Tello, William Isbell and Luis Lumbreras said that Wari, was a synthesis of Warpa, Nazca and Tiahuanaco cultures : a first pre-Hispanic empire,  which assimilate Tiahuanaco agricultural advances and Wari controlled production, being its main challenge to store enough food surpluses. Wari’s leaders planned and developed cities (heads of regions), polychrome pottery with Tiahuanaco iconography and form. Economically they consolidated the ayllus, using heads of regions to religious, political and economic control. They developed the cult to Viracocha under the influence of Pachacamac oracle. Also a fine textilery with alpaca and vicuña threads, accounting and control methods. Speak in favor of an empire, the existence of Wari’s kings, a ruling elite, a warrior nobility, administrative officials in different regions, a priestly class and the presence of artisans, farmers, fishermen, traders, etc.

IV-Wari ended the regionalism, initiating the cultural unification of conquered territories. V- The fast succession of empires (Huari-Inca), suggest a rapid domination and environmental transformation : successful features of empires and civilizations. According to polish archaeologists Milosz Giersz and Paprycja Przadka, the enormous gains of Wari empire caused the decline of smaller cultures within them : Nazca and Mochica with whom the Wari interacted. Makowski, add that between 600 and 1200 years a.c., profound changes occurred in Andean civilization: changes in technologies, ways of building, ways of conceiving the residence of the ruler, dresses, headdresses, ways of weaving, styles of objects, bronze knowledge dissemination, booming production of silver, all originated in the south, winning northern Moche sites. The kero replaces Moche cup, the southern tumi replaces the Northern tumi, changes generated by the expansion of Wari empire, guided by a leader, which agglutinated a lot of people due to its military might. Around 1000 years a.c., Wari empire declined due to inability to meet imperial needs due to serious droughts.

PERU : 2 IMPERIOS PREHISPANICOS

I- Según Krzysztof Makowski (PUCP), los restos arqueológicos funerarios hallados en el Complejo arqueológico El Castillo (Huarmey/Ancash/Peru), son de factura Wari de la sierra, por las caracteristicas de sus cámaras funerarias : pisos enlucidos, muros que se cruzan como un panal de abejas, el empleo de adobe (no piedra), en su construcción, por razones obvias. Los individuos sepultados no están extendidos como los moches, sino sentados en posición wari con envoltorios de pocas vueltas. II-¿Porque el descubrimiento de 63 momias (50 damas de la alta nobleza) y mil 200 objetos (orejeras de plata, collares de oro, pectorales y cerámica de buena calidad), hallados en el Complejo, modificarían la historia prehispanica peruana y la concepción evolutiva de civilizaciones en el Perú y el mundo?. Al estar ubicado El complejo El Castillo en la costa norte peruana, muy lejos de la altiplánica Wari (40 000 habitantes, nucleo urbano amurallado), capital de la cultura Wari (600-1200 años d.c., 25 km al NE de Ayacucho), se solidifica el concepto de Imperio Wari. Makowski, concuerda en que los Huari, fueron un imperio anterior a los incas, dado su modo de conquistar y extenderse. La cultura Wari hunde sus raíces en la tradición Warpa (300 a.d.c.). Las primeras concepciones de civilizaciones prehispánicas (Max Uhle), fueron realizadas en base a macrodescubrimientos arqueologicos : Chavin, Moche, Nazca, Tiahuanaco, Incas, considerados asentamientos independientes, con escasos vasos comunicantes entre ellas.

III-Los recientes hallazgos Wari, confirman una unidad centralizada en la sierra altiplanica, con alerones hacia la costa y selva. Wari, se extendia por el norte hasta Piura, por el sur hasta Cuzco, Arequipa y Tacna y por el E, hasta la ceja de selva (Lamas/San Martin). Julio C. Tello, William Isbell y Luis Lumbreras aseguran que Wari, fue una sintesis de las culturas Warpa, Nazca y Tiahuanaco ; un primer imperio prehispánico, que asimiló los avances agrícolas Tiahuanaco y la producción controlada desde Huari, cuyo principal desafio fué almacenar suficientes excedentes alimenticios. Planificaron y desarrollaron ciudades (cabezas de regiónes), cerámica policroma, con iconografía y forma Tiahuanaco. Económicamente consolidaron los ayllus, usando las cabezas de regiones para el control religioso, político y económico. Desarrollaron el culto a Wiracocha bajo la influencia del oráculo de Pachacamac. Desarrollaron una textileria fina con hilos de lana de alpaca y vicuña, métodos contables y de control. Hablan a favor de un imperio, la existencia de reyes Wari, una elite gobernante, una nobleza guerrera, funcionarios administrativos en diferentes regiones, una clase sacerdotal y la presencia de artesanos, agricultores, pescadores, comerciantes, etc.

IV- Wari puso fin al regionalismo iniciando la unificación cultural de los territorios conquistados. V- La rápida sucesión de 2 imperios (Wari-Inca), sugiere una veloz dominación y transformación del medio ambiente característica de imperios y civilizaciones exitosas. Según los arqueólogos: Milosz Giersz y Paprycja Przadka, las enormes conquistas del Imperio Wari causaron el declive de culturas menores dentro de ellas la Nazca y la Mochica con quienes interaccionaron. Makowski, agrega que entre 600 y 1200 años d.c. ocurrieron cambios profundos en la civilización andina: cambios en tecnologías, modos de construir, formas de concebir la residencia del gobernante, vestidos, tocados, maneras de tejer y teñir, estilos de hacer objetos, difusión de conocimientos del bronce, auge de la producción de plata, originadas todas en el sur, imponiéndose en sitios mochica norteños. El kero reemplaza a la copa moche; el tumi sureño reemplaza al tumi norteño, cambios generados por la expansión del imperio Huari, guiados por un líder, que aglutino a mucha gente dado su poderío militar. Alrededor de 1000 años d.c., se produciría la decadencia y fin del imperio Wari ante la imposibilidad de satisfacer las necesidades, imperiales a consecuencias de graves sequias.
