AGING THERAPY


AGING THERAPY AND PROGERIA

So focused and humble as Ramon y Cajal and Severo Ochoa, the Spaniard biochemist Carlos Lopez Otin (58 years) and his team perform  since   20 years, high-quality research in the Faculty of Medicine of the University of Oviedo (Spain). This note agrees to the foregoing and the huge perspective that generates its last paper  linking the mechanisms of progeria and normal physiological aging with  normal cell reprogramming inhibition,  effect that prevents slow aging or maintaining prolonged youth which by the way is not a  small thing. If Otin and his team continue to explore this niche it  is almost certain that in the coming years, he will be a candidate for the Nobel Prize, given their ideas and youth. Specifies Otin in his most recent paper that in order to prevent cell aging phenomenon it is very  important to facilitate certain normal  reprogramming cell. Otin and his colleagues found that  reconversion  of adult mice  stem cells into induced pluripotent stem cells (iPSCs), is  extremely difficult in old mice in a way  similar to what happens in  progeria and in  normal physiological aging. They further noted that in progeria and in normal aging, the activation of the  protein: NF-kB blocked the generation of iPSCs (embryonic stem cells with  potential for extremely prolonged states of youth) and actívate   the  reprogramming repressor system: DOT1L, reinforcing senescence signals and put off pluripotent genes. Otin concludes that genetic and pharmacological strategies (anti-inflammatory), in order to produce inhibitory action on NF-kB significantly increase the efficiency of reprogramming fibroblasts in cases of Progeria-Guillermo-Nestor and Hutchinson-Gilford  síndromes  and that inhibition of DOT1L extended in vivo half-life and decreases  accelerated aging in mice. Finally, Otin who believes that aging is a disease (cell damage, protein damage, damage to the genome,  epigenome and telomeres) and as such is curable, adds that it is possible to recondition in vitro, damaged cells and then transplant them to a sick  body with   rejuvenation purposes.

 

TERAPIA ANTIENVEJECIMIENTO Y PROGERIA

Tan centrado y humilde como Ramón y Cajal  y Severo Ochoa, el español Carlos López Otin (58 años) y su equipo realizan  desde hace 20 años, investigaciones de gran calidad en la  facultad de Medicina de la Universidad de Oviedo (España). La presente nota se aviene a lo anterior  y a  la  enorme perspectiva que genera su último trabajo en el que relaciona los mecanismos de la progeria y el envejecimiento fisiológico normal  con la inhibición de la  reprogramación celular normal efecto que impide retardar el envejecimiento o mantener una juventud   prolongada lo que por cierto, no  es poca cosa.  Si Otin y su equipo continúan explorando este nicho es casi seguro  que en los próximos años, será candidato al Premio Nobel, dadas sus ideas y juventud.   Especifica  Otin en  su más reciente paper  que  la vejez  impide  la reprogramación celular, facilitando el  envejecimiento. Otin y sus colaboradores descubrieron  que la  reconversión (normal en ratones jóvenes), de células madre de ratones adultos  en  células madre pluripotentes inducidas (iPSCs),   se tornaba extremadamente dificil en ratones viejos en forma semejante a lo que ocurre en la progeria o en el  envejecimiento fisiológico normal. Notaron además   que  la activación de la  proteína : NF-κB  bloqueaba  la  generación de  iPSCs (células madre embrionarias con potencial de estados  de juventud extremadamente prolongada) y activaba  la reprogramación represora :  DOT1L, que refuerza las señales de  senescencia y  desactiva  genes  pluripotenciales. Concluye Otin que las estrategias genéticas y farmacológicas (antiinflamatorios), inhibitorias de la acción del   NF-κB incrementan significativamente la  eficiencia de la  reprogramación de los  fibroblastos del síndrome de Progeria  Néstor–Guillermo y del síndrome de progeria  Hutchinson–Gilford y que la inhibición del  DOT1L  in vivo extiende la vida media y disminuye el envejecimiento acelerado de  ratones gerontes. Finalmente,  Otin quien  opina  que  el envejecimiento es una enfermedad (daño celular, daño  proteico, daño al genoma,   al epigenoma y a los telomeros) y que como  tal es curable, agrega que es  posible reacondicionar in vitro,  células dañadas para luego  trasplantarlas al organismo enfermo con fines de rejuvenecimiento.

Breaktrough Listen



CLOSE ENCOUNTERS OF THE THIRD KIND

Now that the spacecraft: New Horizon allows us to have a close external  and internal view (structure, by analogy) and other data from Pluto, located at the outer limits of our solar system, new avenues are open   for a  large exploration of the  stellar space. We have reached a certain limit, now  we want to see and know more. So far, the following goals: I) The search of similar planets to the Earth with  space probes equipped with energy supply systems (not just, solar), of  higher quality and more duration. II) To create a more realistic spatial mapping, correcting the defects caused by gravity, that  impacting  on light, distorts the actual location of many stellar bodies. III) Intensify the search of  intelligent life beyond our solar system, taking into account  that the Earth is just a point immersed in the interior of  millions of galaxies and billions of billions of stars inside which  carbonaceous compounds exist everywhere, constantly creating organic molecules. The possibility of extraterrestrial life is possible, because  our familiar concepts of physics and chemistry are applied throughout the cosmos. Although it will be relatively easier to find bacterial, plant and perhaps animal life, the most shocking  finding will be to discover intelligent life like ours or perhaps more advanced civilizations. Having just our solar system 5 billion years of existence and the known universe: 15 000 million years, it is likely to raise the existence of intelligent life previous to ours. The encounter with an alien civilization will be a shock and a challenge to our humanity, so it is of utmost necessity, be globally prepared for such a meeting to deal with the consequences of such an event in the best possible way. The search of  intelligent life is not exhausted, because it barely extends about 200 light years, while the unexplored center of the Milky Way is 25,000 light years away from  Earth. If,  there is no life in the Milky Way,  there are  hundreds of billions of other galaxies to explore. Carl Sagan said that life on other planets can be suspected if we see on the surface of them  four biomarkers: greenish by chlorophyll,  water (or similars: liquid methane), carbon and oxygen. Technology has added a fifth technological marker.  The launch of the Project  Breaktrough Listen, by Stephen Hawking and the Russian billionaire Yuri Milner endowed with 100 billion over 10 years to search for intelligent life beyond our solar system, will begin on  December 2015, employing 5 hours daily to capture and interpret repetitive radio waves, TV and laser from the center of the Milky Way to  closest stars of  our solar system, using two of the most powerful existing radio telescopes. The project also strives to promote talks between experts in order to  prepare better for such an encounter inevitably sooner or later. This is an important contribution after the  launch of  the golden phonograph record, attached to the space probes Voyager 1 and 2, Breaktrough Listen, expressed the need of  precise answers to this contingency in all possible areas being an  encounter with bacterial life or with a more advanced civilization. A  civilization similar to ours will compete with us, a  more advanced besides given us an  understanding that our present humanity is just a short episode in our evolution, could help us  and also  submit us to them.

ENCUENTROS CERCANOS DEL TERCER TIPO


Ahora que la sonda espacial : New Horizon nos permite tener  una visión muy cercana de la estructura externa,  interna (por analogía) y    otros datos de  Plutón, ubicado  en  los limites externos de nuestro sistema solar, se abren   nuevas avenidas  para la  exploración in extenso  del  espacio estelar. Hemos llegado a cierto  límite, ahora  queremos ver y saber más. De momento las metas siguientes son: I) Búsqueda de sistemas solares parecidos al nuestro  con sondas espaciales dotadas de  sistemas de aprovisionamiento energético  (no solo, solar), de mayor calidad y duración. II) Crear una cartografía espacial  más real, corrigiendo los defectos ocasionados por la gravedad, la que  al incidir sobre la luz,  distorsiona  la ubicación real de muchos cuerpos estelares. III) Intensificar la búsqueda de vida inteligente mas allá de nuestro sistema solar,  teniendo en cuenta que la Tierra es apenas un punto inmerso entre millones de galaxias y  billones de billones de estrellas en cuyo interior,  compuestos carbonaceos existentes por doquier,  crean constantemente moléculas orgánicas. La posibilidad de vida extraterrestre es factible, en  tanto nuestros   conocidos conceptos de física y química  se aplican en  todo el cosmos. Aunque será relativamente más fácil encontrar  vida bacteriana,  vegetal y quizás  animal,  el hallazgo  más   impactante   será  descubrir  vida inteligente parecida a la nuestra o tal vez civilizaciones más avanzadas. Al tener  nuestro sistema solar apenas   5000 millones de años de existencia y el universo conocido: 15 000 millones de años, es  factible plantear la existencia de vida inteligente  anterior a la  nuestra. El encuentro con una  civilización extraterrestre será un shock y un desafío para nuestra humanidad, por lo que es de suma necesidad, prepararse  globalmente para tal  encuentro  a fin de afrontar las consecuencias de tal evento del  mejor modo posible.  La búsqueda de vida inteligente no está agotada,  apenas  se extiende a unos   200 años luz, mientras el  inexplorado centro  de la Vía Láctea esta a 25,000 años luz de la Tierra. Por lo demás, si   no hay vida en la Vía Láctea puede haberla en cientos de billones de otras galaxias.  Carl Sagan decía que la vida en otros planetas vistas desde la  Tierra tendría cuatro biomarcadores: coloración verdosa por  la clorofila,  presencia de  agua (o similares: metano liquido), carbono y oxigeno. La tecnología ha añadido un quinto marcador, de factura tecnológica. El lanzamiento del Proyecto Breaktrough Listen por parte de Stephen Hawking y el  billonario ruso Yuri Milner dotado  con  100 millones de dólares durante 10 años,  para buscar  vida inteligente mas allá de nuestro sistema solar,  empleara desde  Diciembre del 2015, 5 horas diarias  en captar e interpretar   ondas repetitivas de radio,  TV y laser procedentes del centro de la Vía Láctea y de las  estrellas más cercanas a nuestro sistema solar, empleando 2 de los más potentes radiotelescopios actuales. El proyecto pugna además por   promover conversaciones entre expertos a fin de prepararnos mejor para tal encuentro inevitable más tarde o más temprano. Aparte de ser una importante  continuación al lanzamiento del disco fonográfico de oro,  adherido a las  sondas espaciales  Voyager 1 y 2, Breaktrough Listen,  expresa la necesidad de contar  con respuestas precisas para esta contingencia en todos los ámbitos posibles sea el encuentro con vida  bacteriana o con  una civilización más avanzada.  Una civilización similar  a la nuestra competiría con nosotros, una más avanzada aparte de hacernos entender  que nuestra humanidad actual es apenas  un corto episodio en nuestra evolución,  podría ayudarnos,  pero también someternos.

GENETIC CODE



EXPANDED GENETIC CODE


In 1992, the mathematician E. Sathmary analyzed the consequences of adding or subtracting letters  to the current human genetic code, concluding that although it was possible to increase more characters, there was a risk that the added letters resemble each other, increasing the chances of a mismatch, shape distortion and increased DNA mutagenesis. In contrast, only 2 letters, restrict the emergence of complex bio-organisms. So, the existence of only 4 letters in our genetic code, represent a frozen character, restricted and optimal (fidelity and metabolic efficiency), induced by environmental conditions of the early Earth, where possibly the RNA performed simultaneously storing information and the catalytic work of  proteins. Now, the organic chemist Steven Benner (Foundation for Applied Molecular Evolution. Florida), after 30 years of experiments with artificial genetic codes has added two new letters (p and  z) to the  4  letters of our  genetic code. For Benner, the current DNA is a molecule that would work best, because nucleotides p and  z  align perfectly into  the present DNA, using the same bonds that links  two pairs of current letters, maintaining its natural shape and  because   DNA sequences that contained p and z letters do better than  traditional DNA. While the letter p carries a molecular structure which helps  folding phenomenon, the letter z carries a nitro group, which facilitates the  molecular bonding. According to Benner, an expanded genetic code could generate more combinations, more protein and/or eliminate the necessity of the latter. Instead the system: DNA-RNA-PROTEIN, life on other planets might be evolving with the system: DNA-RNA, capable of evolving faster and transmitting information in both directions. Benner adds that although the current genetic code produces 20 amino acids, a 6-letter code would produce 216  amino acids and billions of proteins, besides to  store far more genetic information. An RNA 6-letter code would do all the work of proteins catalyzing reactions better and produce more complex structures.

CÓDIGO GENÉTICO EXPANDIDO


En 1992, el matemático E. Sathmary analizo las  consecuencias de añadir o restar  letras al código genético humano actual, concluyendo que aunque  era posible incrementar  más caracteres,  existía el riesgo de que las letras añadidas se pareciesen mucho entre sí, incrementándose las posibilidades de un mal apareamiento, distorsión de la forma del DNA e incremento de la mutagénesis. Contrariamente, solo 2 letras,  restringirían  la emergencia de bio-organismos complejos. La existencia de  solo 4 letras en nuestro código genético, representaría un carácter helado, restringido y óptimo (fidelidad y eficiencia metabólica), inducido por  las condiciones  medioambientales de la Tierra primitiva, en las que posiblemente el RNA  realizaba  simultáneamente el almacenamiento   de  información y el trabajo catalítico de las proteínas. Ahora, el químico orgánico Steven Benner (Foundation for Applied Molecular Evolution. Florida),  tras  30 años de experimentos con códigos genéticos artificiales ha  añadido  2 nuevas letras (p y z), a las 4  básicas del  código genético humano. Para Benner, el DNA actual es una molécula que funcionaria mejor de ser perfeccionada, porque  los  nucleótidos p y z se acoplan  perfectamente al DNA actual, empleando los mismos enlaces que unen los 2 pares de letras actuales,  manteniendo su forma natural y porque las secuencias de DNA que contienen letras p y z evolucionan mejor  que el DNA tradicional. Mientras la  letra p porta una estructura molecular que ayuda al plegamiento proteico,  la letra z porta un grupo nitro, que facilita la unión molecular.  Según Benner,  un  código genético expandido generara más combinaciones, más proteínas y/o  prescindirá de estas últimas. En lugar del trío: DNA-RNA-PROTEINAS, la vida en otros planetas podría estar evolucionando con  el dúo: DNA-RNA, capaz de trasmitir  información en ambas direcciones, evolucionando más   rápido. Benner agrega,  que aunque el código genético actual produce  20 amino ácidos, un código de 6 letras produciría  216  amino ácidos y billones de  proteínas, además de almacenar muchísimo más información genética.   Un  RNA de 6 letras haría todo el trabajo de las proteínas, catalizando mejor las reacciones y produciendo estructuras más complejas.

ANTIAGING THERAPIES ARE COMING




SCIENTIFIC BREAKTROUGH OF THE YEAR

For their implications and perspectives, the Dunedin study performed   by researchers from   Duke University (Durham), Otago (New Zealand), California (USA) and London King Collegue that validated  biological age markers in a cohort of young adults,   showed  that some of them were aging more slowly and others more quickly, opens the door to the use of anti-aging therapies (rapamycin and others -successfully employed in experimental animals-), in humans since youth,  being expected  that they can  slow aging, lengthen human lifespan and globally  decrease the  morbidity of late old age. If anti-aging therapies work in humans, the assertion charged to Aubrey de Grey and others who argue that death is a disease and as such can and should be treated will be valid. Dunedin (New Zealand city from where people were recruited) study examined 954 young people (with the same chronological initial age), with 18 physiological biomarkers (HbA1C, cardiorespiratory fitness, waist-hip ratio, FEV1/FVC, FEV1, mean blood pressure, telomere length of leukocyte, BMI, creatinine clearance, urea, lipoproteins, triglycerides, dental health, total cholesterol, white blood cell count, hsCRP, hdl cholesterol, apoB100/apo A1), in 3 points: 26, 32 and 38 years of age, validating two methods by which old age can be measured in young adults, one cross-sectional and other longitudinal, allowing the latter, a quantification of the rate of physiological deterioration in multiple organ systems (lung, periodontal, cardiovascular, renal, hepatic and immune function). These methods were employed in young biological age that still had not developed age-related diseases. At the end of the study (38 years), the biological age varied between 28-61 years, that’s to say,   biomarkers of biological age changed over time and were different in each individual studied. Individuals that aged quickly were   less physically able, showed cognitive decline and brain aging and looked older. The biological age measured in young people could be used to identify and assess causes of aging and rejuvenating therapies.

ACONTECIMIENTO CIENTÍFICO DEL AÑO

Por sus implicancias y perspectivas, el   estudio realizado por investigadores de la Universidades de Duke (Durham),   Otago (New Zeland),  California (USA) y el King Collegue de Londres, al validar   marcadores de edad biológica en una cohorte de adultos jóvenes que mostró que algunos de ellos   envejecían más lentamente y otros más rápido, abre  las puertas al empleo de terapias  antienvejecimiento (rapamicina y otros; empleadas con éxito, en animales de experimentación), a ser empleadas   en humanos desde la juventud, esperándose  enlentecer el envejecimiento,  alargar la vida útil humana y disminuir la morbilidad de la vejez tardía a nivel global. Si  las terapias antienvejecimiento funcionan en humanos, cobrara validez  el aserto de  Aubrey de Grey y otros,  que  sostienen que la   muerte es una enfermedad y que como tal puede y debe tratarse.   El estudio Dunedin (ciudad neozelandesa de donde procedían las personas),  examino a   954 jóvenes (con la misma edad cronológica inicial), con  18 biomarcadores fisiológicos (HbA1C, funcionalidad cardiorespiratoria, relación cintura-cadera, FEV1/FVC, FEV1, media de la presión arterial, longitud del telomero de los leucocitos, BMI, depuración de creatinina, urea, lipoproteínas, triglicéridos, salud dental, colesterol total, cuenta de glóbulos blancos, hsCRP,  colesterol hdl, apoB100/apoA1), en 3 puntos :  26, 32 y 38 años de edad, validándose  2 métodos con  los  que  la vejez  puede ser medida en adultos jóvenes, uno  seccional-cruzado  y otro longitudinal, permitiendo este   último, una   cuantificación de la velocidad del deterioro fisiológico en múltiples órganos y sistemas  (pulmonar, periodontal, cardiovascular, renal, hepático y funcionalidad inmune). Estos métodos  valoraron  la vejez biológica en jóvenes que aun no habían desarrollado enfermedades relacionadas con la vejez. Al finalizar el estudio (38 años),  la edad biológica vario entre 28 a 61 años, es decir  que los biomarcadores de edad biológica  cambiaron con el tiempo y fueron diferentes en cada individuo estudiado. Los individuos que  envejecían  rápidamente se mostraron físicamente menos capaces, mostraron declinación cognitiva y envejecimiento cerebral  y lucían más viejos. La edad biológica medida en jóvenes podría ser empleada usada para identificar causas de envejecimiento y evaluar terapias rejuvenecedoras.

GLUCOSE to MORPHINE



BIOLOGICAL TRANSMUTATION (glucose to morphine)

Since ancient times, alchemists sought to convert (transmute), lead and other elements in gold. Although they did not succeed, in recent times it was discovered that the spontaneous emission of alpha radiation from the isotope: Uranium -238- after several years induces the formation of the stable isotope: lead-206. That's the plot of the elements. But what, in the biological field? Thanks to natural evolution, the things are more or less the same. Recent good or bad news -according to the viewpoint of each one- is that synthetic biologists and biochemists just had isolated the plant gene that induces the conversion of glucose in compounds like codeine, morphine and other drugs. Now, these drugs can be produced by fermenting microbes located within yeast, producing cheaper, safer, with fewer side effects and less addictive properties versions. The problem is that the technique will also be available to drug traffickers. The genes discovered make glucose be converted -through 2 steps- in the intermediate compounds: S-reticulin and R-reticulin that induce the formation of codeine, morphine and the like. Problem: initially microbes produce only small amounts of drugs

TRANSMUTACION BIOLÓGICA (glucosa a morfina)

Desde tiempos muy antiguos, los alquimistas buscaban convertir (trasmutar), plomo y otros elementos en oro. Aunque no lo consiguieron, en épocas más recientes se descubriría que la emisión espontanea de radiación alfa por parte del isotopo: Uranio -238- induce tras varios años, la formación del isotopo estable  plomo-206,  en forma natural. Ese es el terreno de los elementos. Pero ¿y, en el terreno biológico? Gracias a la evolución natural, pasa más o menos lo mismo. La buena o mala noticia, según el punto de vista de cada quien,  es que  biólogos  y bioquímicos sintéticos, acaban de aislar  el gene de la  planta que induce la  conversión de   glucosa  y otros compuestos simples en  codeína, morfina  y otras medicinas. Ahora, estas drogas pueden ser producidas por microbios  fermentadores ubicados  al interior de levaduras, produciendo versiones más baratas, seguras,  con menos efectos colaterales y menos propiedades adictivas. El problema es que la  técnica también estará a disposición de los narcotraficantes. Los genes descubiertos hacen que la glucosa se convierta -mediante 2 pasos- en los compuestos intermediarios: S-reticulina y  R-reticulina,  inductores finales de la formación de  codeína, morfina y similares. Problema: inicialmente los microbios solo  producirán pequeñas cantidades de medicinas  http://news.sciencemag.org/biology/2015/06/final-step-sugar-morphine-conversion-deciphered
