I CAN SEE








BIONIC EYE

Contrary to what the common mortal believes, we do not see with the eyes (these are only intermediaries), but with a certain area of ​​the brain. In the practice 10 people almost blind have begun to see flashes of light, black and white outlines of people standing still and others in motion, this being only the beginning of a novel visual technique. Although the visual perception of flashes of light is instantaneous, it takes some time to interpret these white-dark patterns or in what direction people move. After this, the majority of ex-blind people declare not to feel more alone, conceptuating this technique as of better quality than  genetic or stem cells therapies. It is the 2-Sight Argus® II Retinal Prosthesis System (Argus II/bionic eye), a new techno treatment for severe cases of retinitis pigmentosa and age-related macular degeneration,  the most common causes of blindness. Argus II consists of: 1) a mini-video camera, mounted on: 2) a pair of glasses. Scenes captured by the mini-video camera are sent to: 3) a small computer (video processing unit), located on the outside of the waist, where they are processed and transformed into instructions re-sent to the glasses, via cable, from where they are transmitted wirelessly to: 4) an antenna connected to: 5) receiver  electrode of a chip implanted in the retina (the more pixels of the scene are received by the chip, the better the visual image). 6) From this chip, the signals are re-sent to: 7) an array electrode which emits  small pulses of electricity, which bypass  damaged photoreceptors and stimulate retinal remnant cells transmitting visual information to the brain -along the optic nerve-  creating the perception of light patterns. The first retinal implants were performed at Manchester Royal Eye Hospital (London), at a total cost of € 150,000, per operation.

OJO  BIONICO


Contrariamente a lo que el común de los mortales  cree, no vemos con los ojos (estos son solo intermediarios), sino con cierta área cerebral. En la practica 10  personas casi ciegas  han empezado  a ver   flashes de luz, contornos  en blanco y negro de   personas paradas y otras  en movimiento, siendo esto solo     el principio de una novedosa técnica visual. Aunque la percepción visual de  los  destellos  de  luz  es  instantánea,  tarda algo interpretar  estos claro-oscuros  o,  en qué  dirección se mueven las personas.  Tras ello,    la mayoría  de ex-ciegos  declaran no sentirse más  solos,  conceptuando  a esta   técnica como de mejor calidad que las terapias  genéticas o con  células madre. Se trata del 2-Sight Argus® II Retinal Prosthesis System (Argus II/ojo bionico), un tratamiento de momento circunscrito  a casos severos de  retinitis pigmentosa  y degeneración macular relacionada a la edad,  las causas más comunes de  ceguera. El Argus II,   consta de:1)  una mini-cámara de video, montada sobre: 2) un par de anteojos. Las escenas captadas por la mini-cámara de video son enviadas a: 3) un pequeño  computador  (unidad procesadora del video), ubicada en la parte exterior de la  cintura, donde se procesan y transforman  en instrucciones  re-enviadas a los anteojos, vía cable, de donde son trasmitidas en forma inalámbrica a: 4) una antena conectada al: 5)  electrodo receptor de un chip implantado en la retina (cuanto mas pixeles de la escena sean  recepcionadas por el chip, mejor será la imagen visual). 6) Desde este chip, las señales  son re-reenviadas  a :7) un electrodo emisor,  de  pequeños pulsos de electricidad, que bypasean  los fotoreceptores dañados y estimulan a  células remanentes retinianas  trasmisoras  de  información visual al cerebro -a lo largo del nervio óptico -creando la percepción de  patrones de luz-.  Los primeros implantes retinianos se llevaron a cabo en el Manchester Royal Eye Hospital (Londres), a  un costo total de :150,000,  por operación.

CELL REPROGRAMMING






NEW PROGRESS  in  ANTI-AGING

Normal human embryonic development had a tenet :unidirectional progression/fertilized ovule → adult organism and that during embryogenesis there was an orchestrated process of differentiation tending to generate all  cell types of an adult organism, in a stable environment with minimum cell  and molecular damage. And that,  progressive aging led to a decline in responsible  mechanisms  for adequate cellular health, resulting in a body  inability to maintain normal homeostasis. A concept that begins to change rapidly thanks to a better understanding of aging process. The recent experimental demonstration that mature (differentiated) cells of a living organism can be reprogrammed in vitro and in vivo producing pluripotent cells resembling embryonic stem cells, demonstrates that the process is bidirectional, rather than unidirectional. This was demonstrated by works by Juan Carlos Izpisúa Belmonte et al (In Vivo Amelioration of Age-Associated Hallmarks by Partial Reprogramming, Salk Institute for Biological Studies, San Diego, California). The in vivo reprogramming performed by these researchers in older  mice has revealed a reorganization of DNA methylation and modifications in chromatin, histones and post-translational effects (global epigenetic changes/deregulated remodeling during aging). Similarly, cell reprogramming performed in centenarians and in affects of Progeria Syndrome (premature aging), has allowed to observe elongations of its telomere, rejuvenation of its genetic expression profiles and attenuation of its levels of oxidative stress, generating rejuvenated cells. Surprise : all  program of the gerontologist Aubrey de Gray. Although sometimes the cell reprogramming process performed in mice in vivo, exposed  to  short term (partial reprogramming),  Yamanaka factors (genes: Oct4, Sox2, Klf4 and c-Myc: OSKM), generates teratomas, most times (decreasing the dose of some inducing agents such as doxycycline), it is possible to extend the  life-span of mice  improving  their metabolic diseases, pancreatic and muscular injuries, confirming that aging is the most risk factor for acquiring most diseases. Partial reprogramming rejuvenates cellular phenotypes in human and mouse cells. Another finding of Izpisúa and collaborators is to have determined that  chromosomes carry epigenetic marks, that contribute to maintain tightly united the strands of the DNA and therefore, the activity of the genes. As we age this activity is weakened. As epigenetic modifications are not permanent, it is possible to reprogram adult cells into stem cells, rejuvenating epigenetic marks, slowing the aging process. During aging, there is evidence of damaged DNA accumulation, telomere shortening, cellular senescence, and defects in the nuclear envelope architecture associated to  a unique and universal epigenetic program. A thorough analysis of cellular reprogramming should provide us with a better understanding of the molecular and cellular mechanisms underlying aging  reducing age-related diseases and  improving health and longevity.


ANTIENVEJECIMIENTO: NUEVOS  AVANCES

El desarrollo  embrionario humano normal, tenía un dogma de progresión unidireccional : ovulo  fertilizado → organismo adulto y, que durante la embriogénesis se producía un orquestado  proceso de diferenciación tendiente a generar   todos los tipos celulares   de un organismo adulto, en un medio ambiente estable  con mínimo daño celular y molecular. Y, que el  envejecimiento  progresivo conducía a un declive  de los mecanismos responsables de una adecuada salud celular, dando como resultado una incapacidad  del organismo para mantener la homeostasis. Un concepto que empieza a cambiar rápidamente gracias a una mejor comprensión del proceso de envejecimiento. La reciente demostración experimental de que las células maduras (diferenciadas), de un organismo vivo pueden ser reprogramadas in vitro e in vivo produciendo   células pluripotentes semejantes a células madre embrionarias, demuestra que el proceso es bidireccional, más que  unidireccional. Así, lo demuestran trabajos realizados por Juan Carlos Izpisúa Belmonte  et al (In Vivo Amelioration of Age-Associated Hallmarks by Partial Reprogramming. Salk Institute for Biological Studies, San Diego, California). La reprogramación in vivo realizada por estos investigadores en ratones gerontes, ha puesto en evidencia una reorganización de la metilación del DNA y modificaciones de la cromatina, de las histonas y efectos post-translacionales (remodelación epigenetica global/desreguladas durante  el envejecimiento). Del mismo modo,  la  reprogramación celular realizada en centenarios y en afectos del Síndrome de  Progeria (envejecimiento prematuro), ha permitido observar elongamientos de sus telomeros, rejuvenecimiento de sus perfiles de expresión genética y atenuación de sus niveles de estrés oxidativo,   generando células rejuvenecidas. Todo el programa del gerontólogo  Aubrey de Grey, quien lo creyera.  Aunque algunas veces, la reprogramación celular  realizada en ratones  in vivo, expuestos  a corto plazo (reprogramación parcial), a los factores de Yamanaka (genes :Oct4,Sox2, Klf4 y  c-Myc :OSKM),  genera  teratomas, la mayoría de veces (disminuyendo la dosis de  algunos agentes inductores como la doxiclina), se logra extender la  vida media de los ratones aliviando sus   enfermedades metabólicas, injurias pancreáticas y musculares, confirmándose que el envejecimiento es el factor de mayor  riesgo para adquirir la  mayoria de enfermedades. La reprogramación parcial,  rejuvenece los fenotipos celulares en  células humanas y de ratones. Otro hallazgo de Izpisúa y colaboradores es haber determinado que los   cromosomas portan  marcas epigeneticas, que contribuyen a  mantener   fuertemente unidas las hebras del DNA y por ende, la  actividad de los genes. A medida que envejecemos esta actividad se debilita  Como las modificaciones epigeneticas no son permanentes,  es posible reprogramar las células adultas en células madre,  rejuveneciendo las marcas epigeneticas, enlenteciendo el proceso de envejecimiento. Durante el envejecimiento se ponen en evidencia: acumulación de DNA dañado, acortamiento del telomero,  senescencia celular, y  defectos de la arquitectura de la cubierta nuclear  asociadas a un programa  único y universal epigenetico.  Un análisis prolijo de la  re programación celular nos debe proporcionar   una mejor  comprension de los mecanismos moleculares y celulares subyacentes al envejecimiento disminuyendo las enfermedades relacionadas con la vejez mejorando la salud y  longevidad.

GLIOBLASTOMA

Faced with the obvious difficulties to know personally the Argentinian CONICET (National Council of Scientific and Technical Research of Argentina), we opted for an abstraction of the operation of this Institution based on the critical analysis of an article published by one of its members. I) In a study on the regulation of glioblastoma (brain tumor), led by the biologist Carolina Pérez Castro, we observed that specifically for this research they  worked in collaboration with several national institutions, mostly settled in Buenos Aires, specifically with the IBioBA- Conicet (Institute of Research in Biomedicine of Buenos Aires), FLENI (Foundation for the fight against neurological diseases of childhood, LIAN Laboratory of Research applied to Neurosciences, Laboratory of Molecular Biology, Department of Neuropathology and Molecular Biology (FLENI), Laboratory of Histopathology, Forensic Medical Corps, Supreme Court of Justice, Buenos Aires and other dependencies of Conicet. II) Internationally,  they work as a Partner with the powerful Research Institute: MPSP Max Planck Society of Germany, Linda Crnic Institute for Down Syndrome, Department of Pharmacology, University of Colorado School of Medicine, Aurora, Colorado and the Multigenic Analysis Service, Central Research Unit, Faculty of Medicine, University of Valencia, Spain. III) The research  addresses a current issue, onthe same frontier of neurosciences: mechanisms of origin, properties of self-perpetuation (resembling embryonic stem cells) and growth of glioblastoma, a very aggressive brain tumor. The study underscores the importance of  NSL Chromatin-Modifying Complex Subunit KANSL2  that  regulates Cancer Stem-like Properties of Glioblastoma, being  responsible for regulating the carcinogenic properties (similar to embryonic stem cells), of glioblastoma: self-perpetuation, resistance to therapy and relapse  and  or differentiation.  KANSL2 complex is capable of silencing glioblastoma cells in mouse models. On the other hand, there is a correlation between the levels of KANSL2 and  self-perpetuating activity of glioblastoma, suggesting that the KANSL2 complex would be an excellent candidate as a biomarker of glioblastoma. IV) Thanks to the conception of the study and  multi-institutional support, the paper  was published in the prestigious journal Cancer Research (2016; 76 (18);5383-94).


CONICET   ARGENTINO

Frente a las dificultades obvias para  conocer  personalmente el funcionamiento del CONICET (Consejo Nacional de Investigaciones Científicas y Técnicas), de Argentina,  optamos por   realizar una abstracción del funcionamiento de esta Institución a partir del   análisis crítico  de un artículo  publicado por uno de sus integrantes. I)  En un trabajo en torno  a  la regulación del glioblastoma (tumor cerebral),  liderado por la bióloga Carolina Pérez Castro, observamos que específicamente para esta investigación trabajaron en  colaboración con varias instituciones nacionales, mayormente asentadas en Buenos Aires, específicamente con  el  IBioBA-Conicet (Instituto de Investigación en Biomedicina de Bs Aires), la FLENI (Fundación para la lucha  contra las enfermedades neurológicas de la infancia, el LIAN Laboratorio de Investigación aplicada a Neurociencias,  el Laboratorio de Biología Molecular, Departamento de Neuropatología y Biología Molecular (FLENI), Laboratorio de Histopatología, Cuerpo Médico Forense, Tribunal Supremo de Justicia, Buenos Aires  y otras dependencias del Conicet. II) A nivel internacional trabajan en calidad de Partner  con el poderoso Instituto de Investigación :MPSP (Max Planck Society de Alemania, el  Linda Crnic Institute for Down Syndrome, Department of Pharmacology, University of Colorado School of Medicine, Aurora, Colorado y el Servicio de  Análisis Multigénico, Unidad Central de Investigación, Facultad de Medicina, Universidad de Valencia, España. III) El trabajo de investigación, aborda un tema  de   actualidad, en la misma frontera de las neurociencias:  mecanismos de origen,  propiedades  de autoperpetuacion, semejantes a células madre embrionarias y  crecimiento  del  glioblastoma, un tumor cerebral muy agresivo. El estudio resalta  la importancia de la  subunidad  integral KANSL2  del complejo  no especifico letal (NSL), modificatorio   cromatinico, encargada de   regular  las propiedades cancerígenas (similares a  células madre embrionarias), del glioblastoma :  auto-perpetuación, resistencia a la terapia y recaída de la enfermedad y/o diferenciación. El complejo   KANSL2 es capaz de silenciar a las células del glioblastoma  en modelos de ratones. De otro lado existe correlacion entre los niveles del KANSL2 y la actividad de autoperpetuacion del glioblastoma  sugiriendo que el  complejo KANSL2, sería un excelente   candidato  como  biomarcador del glioblastoma.IV) Merced  a la concepción del estudio y el soporte multiinstitucional   participante,  el artículo fue  finalmente publicado en la prestigiosa revista Cancer Research (2016; 76 (18); 5383-94).

CELLULAR REPROGRAMATION



IN VIVO, ADULT CELLULAR REPROGRAMATION
The application of a previous discovery (S. Yamanaka/2012/Nobel Prize in Medicine: in vitro reprogramming of mature cells into induced stem cells: iPS, by the addition of 4 genes), performed this time in mice, by the spanish researcher Manuel Serrano (National Cancer Research Institute/Madrid), approaches this type of reprogramming to the point where will be possible the self-regeneration of an arm, a leg, a new kidney or directly transform in vivo digestive pancreatic cells into insulin-producing cells, in diabetics. The work of Serrano has allowed us to observe certain novelties: 1) In vivo reprogramming of adult to embryonic cells confirms the existence of totipotential stem cells (more undifferentiated, with presence of placental cells, yolk sac, early hematopoiesis and bone remains), than those obtained by in vitro techniques (without placental cells being  better defined as pluripotential because they are  somewhat more differentiated  than totipotential). 2) Differences that will allow a better understanding of the regulatory molecular signals of cellular reprogramming and the differences between pluripotency and totipotency. Serrano's work leads us to think that ES and iPS cells are slightly more differentiated than totipotentials, which are expected to be more similar to certain undifferentiated, self-reprogrammed cells in vivo contained in amphibian called blastemas that allow the self-generation of its members.

 REPROGRAMACION CELULAR ADULTA, IN VIVO
La aplicación de un  descubrimiento previo (S. Yamanaka/2012/Premio Nobel de Medicina :reprogramación in vitro, de células maduras en  células madre inducidas :iPS, mediante la adición de 4 genes),  realizado esta vez  en ratones in vivo,  por el  español Manuel Serrano (Instituto Nacional de Investigación del Cáncer/Madrid), acerca este tipo de  reprogramación al punto en que será posible  auto-regenerar  un brazo, una pierna, un nuevo riñón o transformar  directamente in vivo  células pancreáticas digestivas  en células productoras de insulina en diabéticos. El trabajo de Serrano ha permitido observar ciertas novedades, a saber : 1) La reprogramación in vivo de células adultas a embrionarias confirma   la existencia de células madre totipotenciales (mas indiferenciadas, con presencia de células placentarias, saco vitelino, hematopoyesis temprana y restos óseos), que  las obtenidas por técnicas  in vitro (sin  células placentarias de por medio,  mejor definidas como pluripotenciales,   algo mas diferenciadas que las totipotenciales). 2) Diferencias que  permitirán  comprender mejor las  señales moleculares regulatorias de la reprogramación celular y las diferencias entre  pluripotencia y totipotencia. El trabajo de Serrano, induce a pensar que las células  ES e iPS son ligeramente más diferenciadas que las totipotenciales, de las que se espera se semejen más a ciertas  células indiferenciadas, auto-reprogramadas in vivo  contenidas en  blastemas de  anfibios que permiten la autoregeneracion de sus  miembros.

QUANTIC BRAIN



A theoretical analysis of the results of behavioral experiments performed in rats (Sechzer JA, Lieberman KW, Alexander GJ, Weidman D and Stokes PE. BiolPsychiatry, 1986; 21 (13): 1258-66), by Matthew Fisher, a theoretical physicist of the University of California, USA, has begun to generate controversy and capture the attention of  international scientific community. If subsequent experiments, to be performed by Fisher and collaborators achieve similar results to those of Sechzer JA, then Fisher could be the next winner of the Nobel Prize in Medicine for opening a new branch of medicine, inductive in turn of a second wave of biological psychiatry. A saga that seems to have been reactivated in 1986, when Fisher devastated by the effects of  a major depression that he suffered decided to follow the track to his own illness. What better then to investigate the mechanisms and behavioral effects of a simple antidepressant: Lithium. Fisher would spend a long time analyzing in depth the effects of 2 isotopes of lithium, chemical entities only different in the number of their nuclear neutrons. The experiment performed by Sechzer JA et al. consisted of administering salts of  2 stable, non-radioactive isotopes of lithium: Li-6 and Li-7 to 3-month-old rats: before, during gestation and during lactation. Li-6- puerperal rats were more affectionate, more cared for, and more frequently breastfed her offspring, while Li-6 and control mothers ignored their offspring and nursed them infrequently. For Fisher these behavioral differences lie in the nuclear spin of each isotope of Lithium, a quantum property that affects the time an atom can remain in entanglement quantum state, isolated from its environment. The smaller the spin of an atom, the lower the interaction of the nucleus with the electric and magnetic fields, having a  quantum decoherence (passage from a quantum entanglement  state  to a classical physical state), slower. By having the Li-6 and Li-7 different numbers of neutrons have different spins. Therefore, Li-7 exhibits too fast decoherence,  for quantum cognition purposes, while Li-6 remains entangled longer, thereby having different behavioral effects.

Hypothesis that suggests a role for quantum mechanisms  in the cognitive process. Fisher believes that the storage of brain quantum information is mediated by phosphorus atoms, which have a half-spin  -a low number- that would make possible long periods of coherence, capable of being further prolonged, if bound to calcium. Fisher thinks Posner molecules (Ca + P) could play the role of a natural brain qbit. After long reflections Fisher (Annals of Physics), thinks that the quantum cellular process begins with a pyrophosphate (2 phosphates released from an ATP). The interaction between the spins of these 2 phosphates causes them to become entangled and can be paired in 4 forms: 3 configurations (triplet/weakly entangled state), which are added to the total spin of one, and a fourth possibility: singlet (maximum entanglement) , capable of producing a zero spin, crucial for quantum computation. Enzymes break   entangled phosphates in 2 free phosphate ions, which remain entangled  even when separated (fast process in singlet state), being able to combine with calcium ions and oxygen becoming  Posner molecules. Neither calcium nor O atoms have nuclear spin to preserve the one-half  total: crucial spin to maintain long periods of coherence, thus protecting the entangled pairs of external interference, maintaining  its consistency for hours, days, weeks, distributed in  long brain distances, influencing the liberation of neurotransmitters and activating neuronal synapses.

CEREBRO CUANTICO

Un análisis teórico de los resultados de experimentos conductuales realizados en ratas (Sechzer JALieberman KWAlexander GJWeidman DStokes PE Biol Psychiatry. 1986;21(13):1258-66), realizado   por  Matthew Fisher, un físico teórico de la Universidad de California, USA, ha empezado a generar  controversia y captar  la atención de la comunidad científica mundial. Si los  experimentos subsiguientes, a ser realizados por Fisher y colaboradores logran  resultados similares a los de  Sechzer JA, entonces Fisher,  podría ser  el próximo  ganador del Premio Nobel de Medicina por   aperturar una nueva rama de la medicina inductora a su vez  de  una segunda gran ola de psiquiatría biológica. Una  saga que parece haberse reactivado   en  1986, cuando Fisher devastado por los efectos de la depresión mayor que padecía  decidió  seguirle  la pista  a  su propia enfermedad. Que mejor  entonces que investigar  los  mecanismos y efectos conductuales de un   antidepresivo simple: el Litio. Fisher pasaría  largo tiempo analizando  de profundis los efectos de 2 isotopos del litio, entes químicos tan solo diferentes en el número de sus neutrones nucleares.  El experimento de Sechzer JA et al, había consistido  en administrar sales de  2 isotopos  estables, no radioactivos de litio: Li-6 y Li-7 a ratas de  3 meses de edad: antes, durante la gestación y durante  la lactancia. Las ratas puérperas  que recibieron  Li-6,  mostraron más afecto, prodigaron más cuidados y amamantaron más frecuentemente a  sus crías, mientras que las que recibieron  Li-7 y las madres controles ignoraron a sus crías y las cuidaron  infrecuentemente. Para Fisher estas diferencias conductuales  radican en el spin nuclear de cada isotopo de Litio,  una propiedad cuántica que afecta al tiempo que un átomo pueda permanecer en estado cuántico entrelazado aislado de su medio ambiente. Cuanto menor es el spin de un átomo,  menor será la interacción del  núcleo con los campos eléctrico y magnético, siendo su decoherencia cuántica  (paso de un estado cuántico entrelazado a un  estado físico clásico),  más lenta. Al tener  el Li-6 y el Li-7 diferente números de neutrones tienen diferentes spins. Por  ello, el Li-7 exhibe una decoherencia,  demasiado   rápida para  propósitos de cognición cuántica,     mientras que el Li-6 permanece  entrelazado  más tiempo, teniendo por ello  diferentes efectos   conductuales.

Hipótesis que sugiere un rol  para los procesos cuánticos en el proceso cognitivo. Fisher  cree que  el almacenamiento de la   información cuántica cerebral es mediada por    átomos de Fosforo, que tienen un spin de una mitad -un numero bajo- que haría  posible tiempos prolongados de coherencia, capaces de ser prolongados  aún más,   si se unen al calcio. Fisher piensa que las moléculas de Posner (Ca +P), podrían hacer el papel de un qbit natural  cerebral.  Tras largas reflexiones Fisher (Annals of Physics), piensa que el proceso celular cuántico se inicia con un  pirofosfato (2 fosfatos liberados de un ATP). La interacción entre los spins de estos  2 fosfatos hace que devengan en  entrelazados, pudiendo aparearse en 4 formas: 3  configuraciones (estado triple/débilmente entrelazado), que se añaden al spin total de uno, y una cuarta posibilidad: singlet (entrelazamiento máximo), capaz de producir un spin zero,   crucial para la computación  cuántica. Las enzimas rompen los fosfatos  entrelazados en 2 iones fosfatos libres,  que continúan permaneciendo  entrelazados  aun cuando estén  separados (proceso veloz  en estado de   singlet), pudiendo combinarse con iones de calcio y oxigeno deviniendo en   moléculas de Posner. Ni el  calcio   ni  los  átomos de O tienen spin nuclear  para preservar el  one-half total:  spin crucial para mantener largos periodos de  coherencia, protegiendo de este modo a los pares  entrelazados   de la  interferencia externa, manteniéndose  la  coherencia  por horas, días, semanas, distribuyéndose por largas  distancias  cerebrales, influenciando la liberación de  neurotransmisores y activando las  sinapsis neuronales.

In many textbooks of Gynecology, it is  explained that women begin to ovulate at age 11, and often quit around 50. With normal menstrual cycles, women ovulate 12 times a year, producing 400 eggs throughout  her  life: about 400 potential children, reduced to 4,  due to   pregnancy and puerperium. Number negligible, because women are born with 400 thousand eggs, which are decreasing to not generate more eggs during menopause. A dogma questioned recently because ovarian biopsies performed to young women who previously received  the chemotherapy regimen ABVD showed areas containing two to four times more eggs than those identified in ovarian biopsies of healthy young women (Evelyn Telfer, University of Edinburgh). Findings that override the limitations to have more children and create   new treatments for  young women with infertility problems and also for older women who want to conceive. Which also  leads us to re-read Brave New World, published in 1932, in which the author Aldous Huxley recreates scenes from the year 2540 BC in which the extinction of natural reproduction and its replacement by another type of artificial  reproduction is anticipated… also in  which thousands of embryos hung on groomed walls  are subjected  to processes  that alter its evolution and development to finish creating: soldiers, workers and of course  leaders. Really a new, brave and challenging world.

UN MUNDO NUEVO Y DESAFIANTE

En diversos libros de  texto de Ginecología se explica   que las mujeres comienzan a ovular  a los 11 años, y que suelen dejar de hacerlo alrededor de los 50. Con ciclos menstruales  normales,  la mujer ovula 12 veces al año, generando 400 óvulos a lo largo de su vida : unos  400 hijos potenciales, reducidos a  4, por las interrupciones propias del embarazo y  puerperio. Numero ínfimo, porque las mujeres nacen con  400 mil óvulos, que van disminuyendo hasta no generar  más, durante la   menopausia.  Un dogma puesto en duda recientemente,  porque  biopsias ováricas realizadas a mujeres jóvenes a quienes se les administro previamente  el régimen  quimioterapico  ABVD, mostraron áreas  conteniendo   2 a 4 veces más óvulos que las identificadas  en biopsias ováricas de  mujeres jóvenes sanas (Evelyn Telfer, University of Edinburgh). Hallazgos que anulan las limitaciones para tener más hijos  y aperturan nuevos tratamientos para mujeres jóvenes con problemas de infertilidad y también para mujeres adultas mayores que desean concebir. Lo que asimismo, nos lleva a releer  Brave New World,  publicada en 1932, en la que el  autor Aldous Huxley  recrea escenas del  año   2540 d C. en las que se anticipa  la extinción de la reproducción natural y su reemplazo     por   otra de  tipo artificial, en la que millares de   embriones colgados en  paredes acondicionadas son sometidos a procesos que alteran  su evolución y desarrollo para terminar creando : soldados, obreros  y  por  supuesto  poderosos líderes.  Realmente un mundo nuevo, bravo y desafiante.

NANOMACHINES




NOBEL PRIZE  OF CHEMISTRY,2016: NANOMACHINES

The Nobel Prize in Chemistry was awarded this year (2016) to the French: Jean-Pierre Sauvage (University of Strasbourg France), because he joined in 1983:  2  ring  molecules of a few nanometers using mechanical links (rather than covalent),  forming  a chain (catenane), in which  one   ring rotated around another by electrical impulses, to: J. Fraser Stoddart (Edinburgh University, UK), for  the   design and  build in 1991 of a  rotaxane: hexagonal molecular ring bonded to a thin wheel  molecular axle. In 1994, Sauvage built an hexagonal ring molecule moving along an  axle emulating muscle movements of  contraction and expansion with  help of  electrical impulses. For its part,  Soddart manage to control   vertical  lifting movements of  a molecular  ring  using electrical impulses and a software connected to a molecule,  and to: Bernard L. Feringa (University of Groningen, Netherlands), for having designed and built  a  rotary molecular  motor  controlled  by   UV light,  capable of performing a task after added  energy and adequate control thanks to added software. In 1999,  Feringa built   another molecular motor with a  blade that continuously rotated in the same direction, the same  that given certain time twirled a glass cylinder 10,000 times bigger   than its nanomotor. Feringa,  also designed a nanocar. It is expected that these molecular machines will be  used for the development of new materials, sensors and energy storage systems. In the near future, these nanomachines will form a new prototype of  robots with the first thinking  robotic brain.

PREMIO NOBEL DE QUIMICA 2016: NANOMAQUINAS

El Premio Nobel de Química fue concedido este año (2016) al francés: Jean-Pierre Sauvage (University of Strasbourg France), por haber unido  en 1983   2 moléculas de unos pocos nanómetros  en forma de anillo empleando enlaces mecánicos (en lugar de covalentes), logrando formar  una cadena (catenano), en la que  un anillo rotaba alrededor del otro mediante  impulsos  eléctricos, a  :J. Fraser Stoddart (Edinburgh University, UK), por diseñar y construir   en 1991, un  rotaxane  :  anillo molecular hexagonal enlazado   a un  delgado  eje de rueda,  molecular (axle). En 1994,  Sauvage  logro  que el anillo hexagonal  se moviese   a lo largo del axle   emulando movimientos musculares de contracción y extensión mediante impulsos eléctricos.  Por su lado Soddart  logro controlar los movimientos  verticales del anillo emulando  movimientos de elevadores empleando impulsos eléctricos y softwares conectados a una  molécula  y  a   Bernard  L. Feringa (University of Groningen, Netherlands), por haber diseñado y construido  motores  moleculares rotatorios  con movimientos controlables  con luz ultravioleta, capaces de efectuar  una tarea al añadirles  energía y controles adecuados merced a  softwares   añadidos. Bernard Feringa desarrollo otro  motor  molecular en 1999 con una hoja rotatoria que  giraba continuamente  en la misma dirección, el mismo  que dado un tiempo hizo  girar un  cilindro de vidrio 10,000 veces más grande que su motor. También diseño un nanocarro. Se espera que estas máquinas moleculares sean   usadas para   el desarrollo de nuevos materiales,  sensores y sistemas de almacenamiento energético. Estas nanomaquinas serán las que en el futuro formen un nuevo  prototipo de robot con el primer cerebro robótico pensante.

TOPOLOGY



NOBEL PHYSICS AWARD, 2016. TOPOLOGY AND PHASES OF MATTER:

The Nobel Prize in Physics was awarded this year (2016) to 3 British settled in American universities: David J. Thouless, F. M. Duncan Haldane and J. Michael Kosterlitz, due to :I) Theoretical discovery of topological transition phases (a world where  matter adopts exotic states). The   topology (study of mathematical properties that change step-wise), was decisive to study unusual phases of matter (superconductors, superfluid, and thin magnetic layers). In 1970, Michael Kosterlitz and David Thouless, overturned   the theory that superconductivity or suprafluidity  not occur in thin layers. Conversely, these scientists argued that superconductivity   will appear  at low temperatures, explaining it by the mechanism of  topological transition phases (superconductivity will disappear  at high temperatures). Later, in 1980, Thouless explain in  electrically  thin layers, that  its conductance could be measured accurately using integers, as these were topological in  nature. II) The discovery of topological phases of matter. In 1980, Haldane discovered that  topological concepts could be used to understand the properties of chains of small magnets in certain materials. Thanks to the above, we now know not only the topological phases of layers and threads, but also  of  3-D ordinary materials. The topology is useful for studying the physics of condensed matter, foreseeing  future applications in materials science, superconductors and in future quantum computers.

El Premio Nobel de Fisica fue otorgado  este año (2016) a 3 británicos afincados en universidades americanas: David J. Thouless, F. Duncan M. Haldane y J. Michael Kosterlitz, por I) El descubrimiento teórico  de las fases de transición topológica (un mundo donde la materia adopta estados extraños). Emplear  topología (estudio de propiedades matemáticas  que cambian  paso a paso), fue  decisivo para  para estudiar fases inusuales de la materia (superconductores, superfluidos, capas magnéticas ultradelgadas). En 1970, Michael Kosterlitz and David Thouless, derribaron  la teoria que afirmaba que la  superconductividad o la  suprafluidez no  ocurrían  en capas delgadas. Contrariamente, estos científicos sostuvieron  que la superconductividad  ocurriría a bajas temperaturas, explicándola mediante  el mecanismo de transición de fases topológica, que hacia desaparecer la superconductividad a altas temperaturas. Más tarde, en 1980, Thouless explicaría  que en  capas eléctricamente  delgadas, su conductancia  podía ser medida en forma precisa empleando  integrales, ya que estas eran de naturaleza topológica. II) El descubrimiento de las fases topológicas de la materia.  En 1980, Haldane descubrió como podían usarse los conceptos topológicos para comprender las propiedades de las cadenas de pequeños magnetos en ciertos materiales. Merced a lo anterior, ahora conocemos las fases topológicas no solo de capas y hebras delgadas, sino también de materiales 3-D ordinarios. La topología es útil para estudiar la física de materia condensada, avizorándose futuras  aplicaciones en ciencia de materiales, superconductores y en futuros computadores cuánticos.

AUTOPHAGY





NOBEL PRIZE IN MEDICINE, 2016: AUTOPHAGY

The Nobel Prize in Physiology or Medicine 2016, was awarded to a professor  of   Tokyo Institute of Technology: Yoshinori Ohsumi, because he elucidated the mechanisms of autophagy, a process that degrades and recycles cellular components, allowing to form embryos  in the most appropriate ways, degraded protein components,  remove bacteria, etc., or, in the case of mutations of genes causing no adequate  autophagy  -not completely degrade  waste proteins-, storing not useful proteins (Alzheimer's disease) or chronic diseases such as diabetes. Although since 1960 was known that cells could destroy their enclosing components in vesicles and transporting them to recycling areas (lysosomes and autophagosomes), it was only in 1990 when Ohsumi using yeast  identified essential genes of autophagy and later discovered similar mechanisms in humans. Autophagosomes can engulf cell contents, damaged proteins and organelles. Ohsumi studied  thousands of yeast mutants, discovering 15 essential genes. Thus it is known today that autophagy is controlled by a cascade of proteins,  each regulating one  stage  of  initiation and formation of autophagosomes. Autophagy control  important physiological functions especially those in which certain cellular components need to be degraded and recycled to provide new energy and cellular building blocks; essential cellular processes against starving  and stress. View higher, autophagy is a mechanism for quality control, critical phase  to counteract the effects of aging. Although  we  ignored  why   the Nobel Prize of  Medicine was awarded  to  a single scientist, we are  happy   to know that  Ohsumi is first  a human being,  a man of very high values, honest, modest, ready to put his own  person  in the last place in  a row of elite scientists.




PREMIO NOBEL DE MEDICINA, 2016: AUTOFAGIA

El premio Nobel  en  Fisiología o,  Medicina, 2016 fue concedido a un profesor del Instituto Tecnológico de  Tokio : Yoshinori Ohsumi,  por haber dilucidado los mecanismos de la autofagia, un proceso que degrada y recicla componentes celulares, permitiendo   dar la forma más adecuada a  embriones en formación, eliminar componentes proteicos degradados de bacterias, etc., o, en el caso de mutaciones de  genes que controlan la autofagia a no degradar completamente restos proteicos innecesarios, almacenando proteínas no útiles (Enfermedad de Alzheimer) o enfermedades crónicas como la diabetes.  Aunque desde 1960 se conocía  que las células podían destruir sus componentes encerrándolos en vesículas y transportándolos a áreas de reciclaje (lisosomas y autofagosomas), fue recién en  1990 en que  Ohsumi,  empleando levaduras logro   identificar los genes esenciales de la autofagia descubriendo más tarde mecanismos similares en humanos.  Los autofagosomas pueden engullir contenidos celulares, proteínas dañadas y organelas. Ohsumi  estudio miles de levaduras mutantes, descubriendo 15 genes esenciales. De este modo se sabe hoy que la  autofagia es controlada por una cascada de proteínas cada una regulando un estadio distinto de iniciación y formación de autofagosomas. La autofagia controla importantes funciones fisiológicas especialmente aquellos en que ciertos   componentes celulares necesitan ser degradados y reciclados a fin de  proporcionar  energía y nuevos bloques de construcción celular ; procesos  celulares  esenciales   frente a la desnutrición y el estrés. Vista a mayor altura, la autofagia es un   mecanismo de control de calidad,  crítico para contrarrestar los efectos del envejecimiento. Aunque ignoramos  porque le dieron el Premio Nobel de Medicina a un solo científico en Medicina, nos contentamos al saber que Oshumi es antes que científico,  un ser humano de dotes muy elevadas,  honesto, modesto, presto a ponerse motu proprio en el último lugar de una fila de científicos de elite.

DESIRED GENES




GENE DRIVES


If CRISPR technique is a molecular biological genomic text processor, that can alter genetic information of any organism; its derivative:  Gene Drives application, seems to have an extraordinary potential to eradicate entire biological species, be a biological weapon  and/or control certain diseases, optimize agriculture, stop the spread of malaria, manage wild ecosystems, etc., introducing desired genetic traits in animals, plants and even human populations. The point is no longer about having programmed children with blue eyes, but to create for example: animals free from all kinds of infectious diseases and so on.  1-The desired gene drive-CRISPR, is inserted into the chromosome of an organism that is sexually mating. 2- The inserted gene drive-CRISPR cleaves a specific portion of the corresponding chromosome from the other parent. 3- Copies of the gene drive-CRISPR of the first parent are made. 4-Then, autoduplicated gene drive-CRISPR restores empty (cut), spaces on both chromosomes, happening the same in each original descendants of the body, ensuring 100% of gene drive transmission to subsequent generations. Although this technique works well on a small scale in fruit flies, mosquitoes and yeasts, it is to be demonstrated that  the introduced desired trait is able to defy the rules of inheritance, in particular: the evolutionary resistance, that will emerge especially in large populations. In order that   gene drive technique accomplish its mission, it is necessary that transmission to the next generations be 100% and not : 50-50% that happened in ordinary conditions. It is argued that once introduced a gene drive it is impossible to reverse ecological damage (if it happened). According to Philipp Messer (Cornell University), only the emergence of natural resistance -after several generations- is the only possibility of eliminating the harmful action of a gene drive. Gene drive application works by replacing (alteration of a specific trait), by which for example a gene drive is able to change the genome of a mosquito, disabling it to carry the malaria parasite. And, by deletion, that is for example: a gene drive would be able to force the offspring of a mosquito to disable all its male reproduction. According to Flaminia Catteruccia (Harvard University), wild populations are able to develop resistance to gene drives, using the option: nonhomologous end-joining rate (cut genomic ends are repaired at random). That is, if the nonhomologous end joining are unchecked, the organism inevitably will develop resistance to scale, initiated somewhere in the genome, which will   spread later, to the surrounding environment. Thus, malaria parasite would be resistant, mutating into a new infectious form in a mosquito previously made resistant by genetic engineering. Lately, Messer has tracing drives gene action in   5000 fruit flies marked with a red fluorescent protein indicator of how far the gene drive action came until the emergence of resistance. Multiple genes are also inserted -instead of a single gene- to create immune   resistance mosquitoes

GENE   DRIVES


Si la técnica   CRISPR, es un procesador molecular  de  textos biológico-genómico- capaz de  alterar la información genética de cualquier organismo, su derivado  : la aplicación Gene Drives, parece tener   la extraordinaria potencialidad de erradicar especies biológicas enteras, ser un arma biológica  y/o,  controlar ciertas enfermedades, optimizar la  agricultura, detener la diseminación de la malaria, manejar ecosistemas salvajes, anular la resistencia a antibióticos, reprogramar genes de mosquitos,etc.,   introduciendo  rasgos genéticos en poblaciones   animales,  vegetales y hasta  de seres humanos. Ya no se trata de tener hijos programados con ojos azules, sino de crear poblaciones animales enteras, libres de todo tipo de enfermedades infecciosas, por ejemplo.  Para ello: 1- Se inserta el  gene drive-CRISPR, en un sitio específico del cromosoma de un organismo que se está apareando  sexualmente.  2-El gene drive CRISPR introducido corta una porción especifica del cromosoma  correspondiente del  otro padre. 3- Se  realizan copias del gen drive del primer padre conteniendo la sección: gene drive-CRISPR. 4-El gene drive  autoduplicado  restaura  los espacios genómicos vacíos (cortados), en ambos cromosomas, asegurando una trasmisión 100% del gen drive a las siguientes generaciones. Aunque esta técnica funciona bien a pequeña escala en la mosca de la fruta, mosquitos y levaduras, el asunto a demostrar es, si el  rasgo deseado introducido  es capaz de desafiar las reglas de la herencia, en especial: la resistencia evolutiva, presta a surgir en grandes poblaciones. Para que la técnica gene drive cumpla su cometido, se necesita  que la  trasmisión  a las siguientes generaciones sea 100% y hasta ahora un rasgo especifico en condiciones ordinarias, solo tiene una chance: 50-50 %, de ser trasmitido. Y, se  aduce que  una vez introducido un gene drive es imposible revertir un daño ecológico (si ocurriese), siendo según Philipp Messer (Universidad de Cornell),  solo la emergencia de  la resistencia  natural -después de varias generaciones- la única posibilidad de eliminar  la acción nociva de un gen drive.  La aplicación gen drive, funciona mediante reemplazo (alteración de un rasgo específico), mediante el cual un gen drive es capaz de  cambiar el genoma de un mosquito, inhabilitándolo por ejemplo a portar el parasito de la malaria. Y,  mediante supresión en la que un gene drive -por ejemplo- sería  capaz de forzar  a la descendencia de un mosquito a inhabilitar la totalidad de su  reproducción masculina.  Según Flaminia Catteruccia (Universidad de Harvard), las poblaciones salvajes son capaces de desarrollar resistencia a  los genes drives, mediante la opción: nonhomologous end-joining rate (extremos genómicos cortados se reparan al azar). Es decir,  si no se  controla  el  nonhomologous end joining, se desarrollara inevitablemente  resistencia a escala, iniciada en algún punto del genoma, que luego se extenderá  al medio ambiente circundante. De este modo, el parasito de la malaria se haría  resistente, mutando  hacia  una nueva forma infecciosa en un mosquito hecho previamente resistente a la malaria mediante ingeniería genética. Últimamente, Messer, ha  rastreando la acción  de gene drives en las cajas de 5000 moscas de la fruta señalizada  con  una proteína fluorescente de color rojo   indicadora de  cuán lejos llego  la acción de los gene drives hasta la emergencia   de  la resistencia. También se  insertan múltiples genes -en lugar de un solo gene drive- para  crear mosquitos   inmunes a la resistencia
